Hashimoto's thyroiditis (HT), the prototype of chronic lymphocytic thyroiditis, is an inflammatory disorder of the thyroid gland characterized by an enlargement of the thyroid (goiter), lymphocytic infiltration of the gland, various degrees of thyroid hypofunction, and circulating antibodies to thyroid antigens. HT is an autoimmune disorder caused by an abnormal immune response to the thyroid gland, both of the humoral and cellular type. The observation that relatives of patients (index cases) are more likely to have the disorder than expected by chance indicates that a genetic predisposition to the disease exists. This view is supported by studies of spontaneous and experimental animal models of the disorder. Several candidate genes have been studied as predisposing factors. This article briefly reviews the evidence for a genetic component in the origin of the disease and summarizes the state of current knowledge on the genes involved.
