Testis tissues from three species of adult cervids were cryopreserved using slow freezing protocol in DMSO or DMSO and FBS. Cryopreserved-thawed testicular tissues were analyzed for the expression of sperm- and spermatid-specific proteins. Retrieved sperm was analyzed for viability, acrosome integrity, morphological preservation, and apoptosis. 20% DMSO was acceptable for cryopreservation of barking deer and 20% DMSO with 20% FBS for sambar and hog deer testes. The study encourages cryobanking of testis of cervids and provides a basis to develop field-friendly USF protocols.
