摘要
目的探寻从麦冬中分离、纯化鲁斯可皂苷元的方法。方法 80%乙醇提取总皂苷,两相酸水解总皂苷,硅胶柱层析分离,甲醇重结晶,薄层色谱(TLC)法、高效液相色谱(HPLC)法检查纯度。结果鲁斯可皂苷元经TLC检查与对照品一致,无杂质斑点,HPLC法检查纯度达98.58%。结论该试验中建立的制备方法简单、安全、可行性强,成本低,效率高,可用于制备鲁斯可皂苷元。
        Objective To investigate the method of separating and purifying ruscogenin from Ophiopogon japonicus. Methods 80%ethanol was used to extract the total saponins,two- phase acid hydrolysis of total saponins,silica gel column chromatography separation,methanol recrystallization; TLC,HPLC methods were used to check the purity. Results Ruscogenin was consistent with the control substance by TLC and no impurity spots. The purity was 98. 58% by HPLC. Conclusion The preparation method is simple,safe,feasible with low cost and high efficiency. The obtained ruscogenin can be used as a reference substance for qualitative and quantitative analysis of traditional Chinese medicine.
