摘要
目的建立梯度洗脱的高效液相色谱法(HPLC)法,测定复方苯甲酸软膏中苯甲酸、水杨酸的含量。方法采用HPLC,色谱柱为Diamonsil C18,以甲醇-0.1%磷酸溶液(45:55)为流动相,波长230nm,流速1.0ml/min,柱温40℃,进行试验。结果苯甲酸、水杨酸分别在2.11~37.98μg/ml和1.99~35.84μg/ml范围内,其与峰面积线性关系较好;精密度、稳定性和重复性实验的相对标准偏差(RSD)均<0.30%;苯甲酸和水杨酸平均加样回收率分别为100.07%和100.01%,RSD分别为0.55%和0.51%。结论 HPLC法测定复方苯甲酸软膏中苯甲酸、水杨酸含量,具有准确、快速、重复性好等优点。
        Objective The contents of benzoic acid and salicylic acid in compound benzoic acid ointment were determined by high performance liquid chromatograph(HPLC) method with gradient elution. Methods HPLC were used,the chromatographic column was Diamonsil C18,the mobile phase was methanol-0.1% phosphoric acid solution(45:55),the detection wavelength was230 nm,the velocity was 1.0ml/min,and the column temperature was 40℃. Results Benzoic acid in 2.11~37.98μg/ml and salicylic acid in 1.99~35.84μg/ml range had good linear relationship with peak area;All the relative standard deviation(RSD),stability and repeatability of the experiment were less than 0.30%;The average recovery rates of benzoic acid and salicylic acid were 100.07%and 100.01%,respectively,and RSD were 0.55% and 0.51%,respectively. Conclusion The HPLC method is used to determine the content of benzoic acid and salicylic acid in compound benzoic acid ointment,and this method has the advantages of accurate,rapid,reproducible.
