乌梅丸对胃肠感染模型小鼠血浆Th1、Th2淋巴细胞因子和胃肠传输功能的影响

摘要
目的:观察乌梅丸对胃肠感染模型小鼠的疗效,同时探讨其对小鼠血浆辅助性T细胞1(Th1)、辅助性T细胞2(Th2)淋巴细胞因子和胃肠传输功能的影响。方法:选取昆明小鼠30只并随机分为空白组、模型组及实验组,各10只。其中模型组及实验组小鼠接受致病性大肠杆菌(EPEC)灌胃制作感染性腹泻模型,空白组用同等剂量生理盐水灌胃。模型制备成功后实验组每日用乌梅丸水溶性生物碱灌胃,剂量按照按人/鼠公斤体重的等效剂量换算,空白组及模型组用同等剂量的生理盐水灌胃,连续干预10 d。比较3组胃排空率、小肠推进率、外周血Th1、Th2淋巴细胞因子表达的变化。结果:模型组及实验组小鼠外周血白细胞计数、中性粒细胞百分比较空白组升高,差异有统计学意义(P <0. 05),其中实验组较模型组降低(P <0. 05)。3组淋巴细胞百分比差异无统计学意义(P> 0. 05)。模型组及实验组小鼠的胃排空率及小肠推进率明显增强,与空白组比较,差异有统计学意义(P <0. 05),其中实验组小鼠的胃排空率及小肠推进率明显较模型组快慢,差异有统计学意义(P <0. 05)。与空白组比较,模型组及实验组小鼠外周血Th2比例、白细胞介素(IL-4)、IL-6、IL-10明显增高,但实验组水平低于模型组,差异有统计学意义(P <0. 05),模型组及实验组Th1比例、IL-2、肿瘤坏死因子-ɑ(TNF-ɑ)、干扰素-γ(IFN-γ)表达较空白组明显升高,但模型组及实验组之间差异无统计学意义(P> 0. 05)。结论:乌梅丸可有效改善胃肠感染模型小鼠的胃肠传输功能,其作用机制可能与介导机体的体液免疫为主。
        Objective: To observe the therapeutic effect of Wumei Pills on gastrointestinal infection model mice,and to explore the effects of Wumei Pills on plasma Th1,Th2 lymphocyte factors and gastrointestinal transport function in mice. Methods: A total of 30 Kunming mice were randomly divided into blank group,model group and control group,with 10 rats in each group. The mice in the model group and the control group were given pathogenic Escherichia coli (EPEC) to simulate infectious diarrhea. The blank group was given the same dose of normal saline. After the model was successfully prepared,the control group was given the watersoluble alkaloids of Wumei Pills daily,and the dosage was converted according to the equivalent dose of human/mouse weight. The blank group and the model group were given the same dosage of saline,and the intervention lasted for 10 days. The gastric emptying rate,intestinal propulsion rate and the expression of Th1 and Th2 lymphocyte factors in peripheral blood were compared among the three groups. Results: 1) The WBC and the percentage of neutrophils in peripheral blood of model group and control group were significantly higher than those of blank group (P < 0. 05),and the WBC and the percentage of neutrophils in control group were lower than those of model group (P < 0. 05). There was no significant difference in lymphocyte percentage between 3 groups (P > 0. 05). 2) The gastric emptying rate and small intestinal propulsion rate of the model group and the control group were significantly higher than those of the blank group (P < 0. 05). The gastric emptying rate and the small intestinal propulsion rate of the control group were significantly higher than those of the model group (P < 0. 05). 3) The ratio of Th2 cells,IL-6,IL-10 and IL-4 in peripheral blood of model group and control group were significantly higher than that of blank group,but the level of experimental group was significantly lower than that of model group (P < 0. 05). The ratio of Th1 cells,the expression of IL-2,TNF-α and IFN-γ in model group and control group were significantly higher than those of blank group,but the level of model group was lower than that of model group (P < 0. 05). There was no significant difference between group and experimental group (P > 0. 05). Conclusion: Wumei Pills can effectively improve the gastrointestinal transport function in mice with gastrointestinal infection,and its mechanism may be mainly mediated by humoral immunity.
