摘要
以‘贵阳小青瓜’(南瓜)未授粉子房为外植体,在离体条件下研究热激时间和TDZ浓度对胚状体诱导的影响。进一步优化试验步骤,探讨直接获得形成再生植株的方法,加速育种进程。结果表明:35℃高温黑暗培养5d后转入25℃光照培养90d能直接形成再生植株;培养基中附加TDZ浓度在0.04~0.06mg/L能诱导离体子房直接产生再生植株,并以0.06mg/L诱导效果最好,出胚率为22.57%,植株再生率为14.30%。
        We used unfertilized ovaries of‘Guiyang xiaoqinggua'(pumpkin)as the explants to study the heat shock time and TDZ concentration on embryo induction via in vitro culture.Furthermore,we optimized the test steps and researched the method of obtaining regenerated plants directly.The results showed that the best obtained pumpkin plants were inducted regenerated plants directly under dark at 35℃for 5 d,and then moved to light condition at 25℃for 90 d;The unfertilized ovaries can induct regenerated plants in the TDZ concentration at 0.04-0.06 mg/L,the best TDZ concentration was 0.06 mg/L,and the ratio of inducted embryo and regenerated plants were 22.57% and 14.30%,respectively.
