平喘宁调节ERK信号通路相关蛋白Ras、MEK1、C-FOS干预哮喘气道重塑的效应机制研究

摘要
目的:研究平喘宁对寒哮大鼠肺组织中ERK信号通路中Ras、MEK1、C-FOS等相关蛋白表达水平的影响,探讨平喘宁干预哮喘气道重塑的作用机制。方法:SPF级雄性大鼠140只,随机分为正常组、模型组、地塞米松组、桂龙咳喘宁组以及平喘宁高、中、低剂量组。4周后采用HE染色观察肺组织病理形态学变化;采用Western Blot法检测大鼠肺组织Ras、MEK1、 C-FOS的蛋白表达。结果:通过HE染色后可以发现,与正常组比较,模型组有明显气道重塑现象;与模型组比较,各治疗组气道重塑均得到适当缓解。通过Western Blot法检测发现,与正常组比较,模型组Ras、MEK1、C-FOS等相关蛋白的表达均有增高(P<0.01);而与模型组比较,除平喘宁小剂量组在下调ERK1指标的表达上效果不明显外,其余各治疗组Ras、MEK1、C-FOS等相关蛋白的表达均显著下降(P<0.01,P<0.05)。结论:平喘宁可通过调节ERK信号通路,明显减轻哮喘大鼠气道炎性反应,延缓气道重塑。
        Objective: To investigate the effects of Pingchuanning on the expression of Ras, MEK1, C-FOS and other related proteins in ERK signaling pathway in lung tissue of rats with cold asthma, and to explore the mechanism of Pingchuanning intervention in asthmatic airway remodeling. Methods: A total of 140 SPF male rats were randomly divided into normal group, model group, dexamethasone group, Guilong Kechuanning group and Pingchuanning high, medium and low dose groups. After 4 weeks, the histopathological changes of lung tissue were observed by HE staining. The protein expression of Ras, MEK1 and C-FOS in rat lung tissue was detected by Western Blot. Results: After HE staining, it was found that the model group had significant airway remodeling compared with the normal group; compared with the model group, airway remodeling was appropriately relieved in each treatment group. Western Blot analysis showed that compared with the normal group, the expressions of related proteins such as Ras, MEK1 and C-FOS were increased(P<0.01). Compared with the model group, except for the low dose of Pingchuanning group was not obvious, the expression of Ras, MEK1, C-FOS and other related proteins in the other treatment groups were significantly decreased(P<0.01, P<0.05). Conclusion: Pingchuanning can significantly reduce airway inflammatory response and delay airway remodeling by regulating ERK signaling pathway.
