益气生骨颗粒促进大鼠骨折愈合的实验研究

摘要
目的:观察益气生骨颗粒对骨折愈合的促进作用。方法:选用32只3月龄大鼠,全部手术制作左侧桡骨中段1mm骨缺损的骨折模型,随机分为用药组和模型组,每组16只。造模次日,用药组予益气生骨颗粒,12 g/(kg·d),分2次灌胃;模型组予白开水,2 mL/次,2次/d。于造模后第7、14、21、28天分批处死动物,取左侧(造模侧)骨痂先拍X线观察,然后再作组织学观察;右侧(健侧)尺骨行生物力学性能检验。结果:骨痂X线观察显示用药组在14、21 d骨痂量多、密度高且缺损基本填满,而模型组多个样本缺损尚未填满、骨痂量少、密度浅淡。组织形态学观察显示造模后7 d时,用药组与模型组均无血肿存在,成纤维细胞量很少;用药组骨痂在14、21、28 d均比模型组更成熟,表现为骨小梁排列更加规则。在21、28 d,骨性组织面积高于模型组(P<0.05)。生物力学测试,健侧尺骨的载荷-位移曲线在第2周低于模型组,而在第1、3、4周两组比较,差异无统计学意义(P>0.05)。结论:益气生骨颗粒能够促进骨折愈合,表现为促进骨痂的生长,提高骨痂的钙化质量。
        Objective: To observe the effect of Reinforcing Qi and Producing Bone Granules(益气生骨颗粒)on fracture healing. Methods: The thirty-two SD rats aged three months were randomized into two groups of 16 each: the model group and the therapy group. All of them were performed into left-radial 1 mm bone coloboma model on operation. The next day of operation, they were fed orally with boiled water(2 mL every time, twice a day) and "Reinforcing Qi and Producing Bone" Granules 12 g/(kg·d), twice a day. On the 7 th, 14 th, 21 st, 28 thday after operation, the animals were scarified, left radius were used to measure X-ray films and examined histomorphological of bone callus, right ulna were used to measure the biomechanics. Results: Bone callus macroscopic X-rays reveal that compared with the model group, on the 14 th, 21 stday after operation the therapy group have more of bone callus, higher density and its deficits are almost fulfilled. Tissue-morphism reveals that on the 7 thday after operation, the model group and the therapy group both have no hematoma, and have small number of fibroblasts. And bone callus of the therapy group are of ripenss much more, the array of bone trabecula are more regular, and the area of osteoid tissue in therapy group is bigger on 21 st, 28 thday(P<0.05). The biomechanics reveal that in the therapy group, the curve of download-displacement of right ulna is lower on 14 th day than that of the model group(P<0.05). Two groups have no notable distinction with each other on the 7 th,14 th, 21 st, 28 thday(P>0.05). Conclusions: "Reinforcing Qi and Producing Bone" Granules could surely promote fracture healing, and it can promote the growth of bone callus and improve the quality of calcification of bone callus.
