摘要
地木耳为原料,采用单因素和响应面法优化了地木耳多糖的热水提取工艺,且进行脱蛋白和醇沉,得到地木耳多糖并对其理化性质进行测定。实验结果表明,响应面优化最佳工艺条件为料液比为1/55.62 g/mL,提取温度为366.37 K,提取时间为196.73 min,预测多糖得率为10.76%。实际提取条件修正为液比为1/50 g/mL,提取温度为370.15 K,提取时间为190 min,根据修正条件进行提取,多糖得率为10.43%。用硫酸钡比浊法测得地木耳多糖中硫酸根含量为2.18%;糖醛酸含量为33.41%,此地木耳多糖为酸性多糖。研究结果可为地木耳多糖提取工艺研究提供科学依据。
        Nostoc commune was used as raw material, and the hot water extraction process of polysaccharide from Nostoc commune was optimized by using single factor and response surface method. After deproteinized and alcohol precipitation, the polysaccharides from Nostoc commune were obtained and their physicochemical properties were determined. The experimental results showed that the optimal reaction condition for response surface optimization was 1/55.62 g/mL of solid-liquid ratio, 366.37 K of extraction temperature, and 196.73 min of extraction time, and the predicted polysaccharide yield was 10.76%. The actual extraction conditions were corrected to the solid-liquid ratio of 1/50 g/mL, the extraction temperature of 370.15 K, and the extraction time of 190 min. According to the amended conditions for extraction, the polysaccharide yield was 10.43%. The content of sulfate radical in Nostoc commune polysaccharide was 2.18% with barium sulfate turbidimetry. The uronic acid content was 33.41%, indicating that the polysaccharide of this Nostoc commune was acidic polysaccharide. The results could provide scientific reference for the extraction of polysaccharide from Nostoc commune.
