三种质量浓度医用臭氧局部注射对兔膝骨性关节炎行为学及滑膜匀浆中IL-1、TNF-α水平的影响

摘要
目的探讨三种质量浓度臭氧对新西兰兔膝骨性关节炎行为学及血清中致炎因子IL-1、TNF-α水平的影响。方法随机将90只新西兰兔分成5组,分别为正常组、模型组、臭氧20μg/ml组、臭氧30μg/ml组、臭氧50μg/ml组,正常组不进行任何处理,模型组仅造模,其余3组造模之后注射3 ml的臭氧,臭氧治疗1 d、5 d和10 d后,观察各组新西兰兔Lequesne MG膝关节评估级别及滑膜匀浆中IL-1、TNF-α的水平变化情况。结果 Lequesne MG膝关节评估在局部反应、步态反应、关节活动和关节肿胀方面各个臭氧治疗组与模型组、正常组相比差异有统计学意义(P<0.05);治疗1 d、5 d、10 d后,IL-1及TNF-α两种因子的质量浓度变化一致,均为臭氧50μg/ml组>模型组>臭氧30μg/ml组>臭氧20μg/ml组>正常组;臭氧20μg/ml组、臭氧30μg/ml组的IL-1、TNF-α的质量浓度均显著降低(均P<0.05);臭氧治疗10 d后,臭氧20μg/ml组兔滑膜匀浆中IL-1、TNF-α质量浓度与正常组相比差异不显著(P>0.05);臭氧50μg/ml组的IL-1、TNF-α的质量浓度高于模型组(均P<0.05)。结论臭氧浓度为20、30μg/ml时可以显著改善新西兰兔行为学功能,降低新西兰兔膝关节损伤部位滑膜匀浆中IL-1、TNF-α的质量浓度,延缓KOA病程的进展;当臭氧浓度为50μg/ml时加重了新西兰兔骨关节的损伤。
        Objective To investigate the effects of three concentrations of ozone on the expressions of IL-1 and TNF-αin osteoblastic arthritis of New Zealand rabbits. Methods Ninety New Zealand white rabbits were randomly divided into five groups:normal group,model group,20 μg/ml ozone group,30 μg/ml ozone group and 50 μg/ml ozone group. The last three groups were injected with 3 m L of ozone. After ozone treatment for 1 d,5 d and 10 d,the changes of Lequesne MG knee joint grade evaluation and levels of IL-1 and TNF-α in synovial homogenates in each group were observed. Results There was significant difference between the model group and normal group in local reactions,gait reaction,joint activity and joint swelling( P<0. 05) according to Lequesne MG knee evaluation. After 1 d,5 d and 10 d,the concentrations of IL-1 and TNF-αin the two groups changed in the same way: ozone 50μg/ml group>model group>ozone 30 μg/ml group>ozone 20 μg/ml group>normal group. The expressions of IL-1 and TNF-αin ozone 20 μg/ml group and 30 μg/ml ozone group were significantly decreased( all P<0. 05). After 10 days of ozone treatment,the concentrations of IL-1 and TNF-αin rabbit synovial homogenates of the 20 μg/ml ozone group were significantly decreased. Compared with the normal group,the difference was not significant( P >0. 05). The concentrations of IL-1 and TNF-α in the 50 μg/ml ozone group was higher than that in the model group( all P<0. 05). Conclusion Ozone at the concentrations of 20 μg/ml and 30 μg/ml can significantly improve the behavioral function of New Zealand rabbits,reduce the concentration of IL-1 and TNF-αin their synovial homogenates,delay the progression of KOA. However,ozone at a concentration of50 μg/ml can aggravate bone and joint damage of New Zealand rabbits.
