摘要
为了探讨猪肌苷酸含量与相关基因表达的相关性,试验采用高效液相色谱法测定宁乡猪杂交组合杜×宁、巴×宁、巴×巴宁和巴宁×巴宁背最长肌肌苷酸含量,采用实时荧光定量PCR(qPCR)技术检测背最长肌ADSL、GPAT和PURH基因的表达水平。结果表明:巴×宁猪肌苷酸含量显著高于巴×巴宁猪(P<0.05);ADSL基因在巴×宁猪中的表达量极显著高于巴×巴宁猪(P<0.01),GPAT和PURH基因在巴×宁猪中的表达量均显著高于巴×巴宁猪(P<0.05);ADSL和GPAT基因表达量与肌苷酸含量均呈显著正相关(P<0.05),PURH基因表达量与肌苷酸含量呈极显著正相关(P<0.01)。说明ADSL、GPAT和PURH基因与肌苷酸沉积密切相关,可作为猪肉品质的选择依据。
        To explore the correlation between inosine monophosphate( IMP) contents and related gene expressions in pigs,high performance liquid chromatography method was used in this experiment in determining IMP contents of longissimus dorsi in crossbreed pigs including Duroc×Ningxiang,Berkshire×Ningxiang,Berkshire×( Berkshire×Ningxiang),( Berkshire×Ningxiang) ×( Berkshire×Ningxiang),and real-time quantitative PCR method was employed in detecting the expression levels of ADSL,GPAT and PURH genes. The results showed that IMP content in Berkshire×Ningxiang was significantly higher than that in Berkshire×( Berkshire×Ningxiang)( P<0. 05). The expression level of ADSL gene in Berkshire×Ningxiang was very significantly higher than that in Berkshire×( Berkshire×Ningxiang)( P< 0. 01). The levels of GPAT and PURH genes in were Berkshire × Ningxiang were significantly higher than those in Berkshire ×( Berkshire × Ningxiang)( P < 0. 05). A significantly positive correlations( P<0. 05) between expression levels of ADSL and GPAT genes and IMP contents were obtained,and a very significantly positive correlation( P<0. 01) between expression level of PURH gene and IMP contents was received. These results suggested that ADSL,GPAT and PURH genes are closely related to IMP deposition,which could serve as a basis for the selection of meat quality.
