白藜芦醇在胰腺癌大鼠中的应用效果及对T细胞免疫增强效应的影响研究

摘要
目的探讨白藜芦醇在胰腺癌大鼠中的应用效果及对T细胞免疫增强效应的影响。方法选择2017年6月至2017年12月参与实验的SD大鼠30只作为对象,随机取SD大鼠10只,设为空白对照组;其余20只大鼠通过胰腺被膜下包埋二甲基苯蒽(DMBA)建立胰腺癌大鼠模型,建模成功后随机分为白藜芦醇组(n=10)和5-Fu尿嘧啶组(n=10)。空白对照组不采取有效的措施干预,5-Fu尿嘧啶组静脉注射5-Fu尿嘧啶12 mg/kg,白藜芦醇组灌胃给药白藜芦醇水溶液100μg/ml。比较三组肿瘤体积、肿瘤重量、T细胞免疫水平及HE染色。结果白藜芦醇组肿瘤体积与肿瘤重量低于5-Fu尿嘧啶组(P <0. 05); 5-Fu尿嘧啶组肿瘤体积与肿瘤重量,高于空白对照组(P <0. 05);白藜芦醇组CD3+、CD4~+及CD4~+/CD8~+水平,均高于5-Fu尿嘧啶组(P <0. 05),低于空白对照组(P <0. 05);白藜芦醇组CD8+水平,低于5-Fu尿嘧啶组(P <0. 05),高于空白对照组(P <0. 05); HE染色结果表明:白藜芦醇组瘤体体积较小,肿瘤细胞分布均匀、排列致密,分裂像相对较少,胞浆及细胞间质均较少; 5-Fu尿嘧啶组HE染色结果表明瘤体体积较大,肿瘤细胞分布不均匀、排列紊乱;空白对照组均为胰腺组织,未见癌细胞。结论白藜芦醇用于胰腺癌大鼠中能缩小肿瘤体积及肿瘤重量,提高T细胞免疫水平,促进肿瘤细胞凋亡。
        Objective To investigate the effect of resveratrol in pancreatic cancer rats and its effect on T cell immune enhancement.Methods 30 SD rats were enrolled from June 2017 to December 2017. 10 SD rats were randomly selected as blank control group. The remaining20 rats received phenyl hydrazine( DMBA) submergence for pancreas. After successful modeling,they were randomized into resveratrol group( n= 10) and 5-Fu uracil group( n = 10). The blank control group was without effective intervention. The 5-Fu uracil group was intravenously injected with 5-Fu uracil 12 mg/kg,and the resveratrol group was intragastrically administered with 100 μg/ml resveratrol solution. Comparison regarding tumor volume,tumor weight,T cell immunity level and HE staining was made among all groups. Results Tumor volume and tumor weight of resveratrol group were lower than those of the 5-Fu uracil group( P < 0. 05). Tumor volume and tumor weight of 5-Fu uracil group were higher than blank control group( P < 0. 05). The levels of CD3~+,CD4~+and CD4~+/CD8~+in resveratrol group were higher than in 5-Fu uracil group( P < 0. 05) but was lower than in blank control group( P < 0. 05). The level of CD8~+in resveratrol group was lower than in 5-Fu uracil group( P < 0. 05) but was higher than in blank control group( P < 0. 05). HE staining of resveratrol group turned out relatively small tumor volume with evenly distributed and densely arranged tumor cells,less cytoplasm and intercellular substances and were rarely in split phases.HE staining of 5-Fu uracil group turned out larger tumor volume with unevenly distributed and disorderly arranged tumor cells. HE staining of blank control group showed normal pancreatic tissue. Conclusion Resveratrol can inhibit pancreatic tumor while increase T cell immunity and promote tumor cell apoptosis in rats.
