清肠解毒汤对溃疡性结肠炎湿热证大鼠结肠黏膜病理组织学改变及NF-κBp65表达影响

摘要
目的:分析清肠解毒汤对UC湿热证大鼠结肠黏膜病理组织学改变及核因子κB(NF-κB)p65表达影响,为临床患者诊疗提供一些实验室依据。方法:选取SPF级Wistar雄性大鼠45只,随机数字表法将分成三组,正常组、模型组和观察组,每组各15只,观察组和模型组制备UC湿热证大鼠模型,观察组大鼠使用清肠解毒汤灌胃,剂量为0.7g/kg,正常组和模型组大鼠使用同剂量生理盐水灌胃,每天1次,共灌胃14天。末次给药后各组大鼠禁食24小时,而后称量大鼠体重,腹腔注射剂量为0.8ml/100g的2.4%水合氯醛麻醉,腹腔剖开后选取结肠组织,常规行HE染色,在光镜下查看各组大鼠肠黏膜组织的病理状况,同时依据结肠病理组织的评分准则对大鼠进行评分,免疫检测大鼠肠黏膜组织内IL-6、NF-κBp65及IL-8蛋白表达状况。结果:模型组大鼠体重比正常组下降,病理评分比正常组上升,观察组体重比模型组上升,病理评分比模型组下降,差异均有统计学意义(P<0.05);模型组大鼠IL-6、NF-κBp65及IL-8蛋白表达量比正常上升,观察组IL-6、NF-κBp65及IL-8蛋白表达量比模型组下降,差异均有统计学意义(P<0.05)。结论:清肠解毒汤可显著改善湿热证UC大鼠结肠黏膜组织的病理状态,降低黏膜组织内L-6、NF-κBp65及IL-8蛋白表达量。
        Objective:To analyze the effect of Qingchang Jiedu Decoction on the histopathological changes of colonic mucosa and the expression of nuclear factor B(NF-B) p65 in UC damp-heat syndrome rats,and provide some laboratory basis for the diagnosis and treatment of clinical patients.Methods:A total of 45 male Wistar male rats of SPF grade were randomly divided into three groups:normal group,model group and observation group,with 15 rats in each group.The rats in the observation group and the model group were prepared with UC damp-heat syndrome.Rats in the group were treated with Qingchang Jiedu Decoction at a dose of 0.7 g/kg.Rats in the normal group and the model group were intragastrically administered with the same dose of normal saline once a day for 14 days.After the last administration,the rats in each group were fasted for 24 hours,then the body weight of the rats was weighed,and the intraperitoneal injection was 0.8 ml/100 g of 2.4% chloral hydrate.The colon tissue was selected after abdominal dissection,and HE staining was performed routinely.The pathological conditions of intestinal mucosa in each group were observed under light microscope.The rats were scored according to the scoring criteria of colonic pathology.The expressions of IL-6,NF-(Bp65 and IL-8 in intestinal mucosa of rats were detected by immunoassay.Results:The body weight of the model group was lower than that of the normal group.The pathological score was higher than that of the normal group.The weight of the observation group was higher than that of the model group,and the pathological score was lower than that of the model group.The difference was statistically significant(P<0.05).The expression levels of IL-6,NF-(Bp65 and IL-8 protein of the model group increased normally.The expression levels of IL-6,NF-(Bp65 and IL-8 in the observation group were lower than those in the model group,and the difference was statistically significant(P<0.05).Conclusion:Qingchang Jiedu Decoction can significantly improve the pathological state of colonic mucosa in MSC rats with damp-heat syndrome and reduce the expression of IL-6,NF-Bp65 and IL-8 protein in mucosa.
