三七总皂苷对过氧化氢所致HUVECS损伤的保护作用

摘要
目的:探讨三七总皂苷(血塞通)从抗氧化途径对人脐静脉内皮细胞(HUVECS)的保护作用。方法:体外培养HUVECS细胞,采用CCK8法,不同剂量血塞通对正常细胞干预,然后得到25、50、100mg/L为最终剂量。通过不同剂量过氧化氢对于HUVECS的损伤,CCK8法检测细胞活性,得出500μmol/L,3h确定造模条件。按照对照组、模型组、三七总皂苷(血塞通)高、中、低剂量五个组展开实验,对LDH、SOD、CAT、NO、NOS相关氧化指标进行检测。结果:三七总皂苷(血塞通)对正常细胞在0~100mg/L具有促进生长的作用,超过此剂量具有抑制作用;在促进范围剂量内可以降低LDH、MDA表达,升高SOD、CAT、NO、NOS表达。结论论:三七总皂苷(血塞通)对HUVECS氧化而产生的损伤具有保护作用,其机制可能是通过抗氧化途径保护细胞。
        Objective: Study PNS(Xuesetong) from the anti-oxidative pathway in human umbilical vein endothelial cells(HUVECS) protection.Methods: HUVECS cells were cultured in vitro, using CCK8 method, different doses PNS(Xuesetong) intervention on normal cells, then get 25, 50,100 mg/L for the final dose. By different doses of hydrogen peroxide for HUVECS damage, CCK8 cell activity assay, draw 500μmol/L, 3 h modeling to determine the conditions. In accordance with the control group, model group, Xuesaitong high, medium and low dose group launched five experiments. Of LDH, SOD, CAT, NO, NOS related oxidative indexes were detected. Results: PNS(Xuesetong) normal cells in the 0~100 mg/L can promote the growth of more than this dose inhibited. In promoting the dose can be reduced in the range of expression of LDH, MDA, elevated SOD,CAT, NO, NOS expression. Conclusions: PNS(XST) injury HUVECS oxidation and has a protective effect, the mechanism is possible to protect cells by anti-oxidative pathway.
