血清miRNA-21、IL-25表达水平与肾癌患者临床特征的关系

摘要
目的探讨血清微小RNA-21(miRNA-21)、白细胞介素-25(IL-25)表达水平与肾癌患者临床特征的关系。方法选取60例肾癌患者作为肾癌组,选取60例健康体检者作为对照组。采用酶联免疫吸附测定(ELISA)法检测两组受试者的血清IL-25表达水平,采用逆转录聚合酶链反应(RT-PCR)技术检测两组受试者的血清miRNA-21表达水平。分析不同Fuhrman分级、肿瘤直径、肿瘤分期、淋巴结转移情况的肾癌患者血清miRNA-21、IL-25表达水平的差异。结果肾癌组患者的血清miRNA-21、IL-25表达水平均明显高于对照组(P<0.01);不同Fuhrman分级、肿瘤直径、肿瘤分期、淋巴结转移情况肾癌患者的血清miRNA-21表达水平比较,差异均无统计学意义(P>0.05)。肿瘤分期为Ⅲ～Ⅳ期、有淋巴结转移肾癌患者的血清IL-25表达水平高于肿瘤分期为Ⅰ～Ⅱ期、无淋巴结转移的患者,差异均有统计学意义(P<0.05)。结论肾癌患者的血清miRNA-21、IL-25表达水平较健康人群明显升高,且血清IL-25水平升高可能与肾癌患者的病理学分期和淋巴结转移情况有关。
Objective To investigate the relationship between the expression of serum microRNA-21(miRNA-21) and interleukin-25(IL-25) and the clinical characteristics of renal carcinoma patients. Method A total of 60 patients with renal carcinoma were included as renal carcinoma group, and another 60 healthy subjects were enrolled as control group. The serum IL-25 level in both groups were detected by enzyme-linked immunosorbent assay(ELISA), and the level of miRNA-21 in both groups were determined using reverse transcription polymerase chain reaction(RT-PCR). The differences of the expression of serum miRNA-21 and IL-25 in renal carcinoma patients with different Fuhrman classification, lesion diameter, tumor stage, and lymph node metastasis were analyzed. Result The serum levels of miRNA-21 and IL-25 in the renal carcinoma group were significantly higher than those in the control group, and the differences were statistically significant(P<0.01); though there were no evident differences observed regarding the serum miRNA-21 expression among renal carcinoma patients with varying Fuhrman classification, tumor diameters, tumor stages, or lymph node metastasis(P>0.05). Significantly higher serum IL-25 levels were noted in renal carcinoma patients with tumor stage Ⅲ-Ⅳ and lymph node metastasis compared with those with tumor stage Ⅰ-Ⅱ and no lymph node metastasis, demonstrating statistically significant difference(P<0.05). Conclusion The expression of serum miRNA-21 and IL-25 in patients with renal carcinoma are significantly higher than those in normal population, the elevated serum IL-25 level may be related to the pathological stage and lymph node metastasis in renal carcinoma patients.
