摘要
为探究大蒜中抗病毒的活性成分二烯丙基二硫化物(Diallyl disulfide,DADS)在体内抗鸡马立克氏病毒(Marek's Disease Virus,MDV)的效果,以弱毒疫苗株CVI988/Rispens接种鸡胚成纤维细胞(CEFS)获取大量的MDV,感染雏鸡建立模型。主要通过病理组织切片的观察、血清生化指标的检测以及实时荧光定量QPCR等方法,探讨DADS在鸡体内抗鸡马立克氏病毒的有效浓度范围以及作用效果。结果表明:DADS浓度为250,500,750μmol/m L时能有效降低雏鸡血清中天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)以及甘油三酯(TG)的水平,同时也有降低病毒拷贝数的作用。所以3种浓度的DADS能在疾病早期有效抑制患马立克氏病鸡肝脏肿瘤生长并具有体内抗病毒的效果。
        The effect of Diallyl disulfide( DADS),an active ingredient of anti-virus in garlic,on against Marek' s Disease Virus( MDV) in vivo were investigated. Large amounts of MDV were obtained by inoculating chicken embryo fibroblasts( CEFS) with attenuated vaccine strain( CVI988/Rispens) o.A model was established by infecting chicks with the virus. The effective concentration range and effect of DADS against chicken Marek's virus in vivo determined based on the methods such as observation of pathological tissue sections,detection of serum biochemical markers,and real-time fluorescent quantitative Q-PCR. The results showed that levels of aspartate aminotransferase( AST),alanine aminotransferase( ALT),and triglyceride( TG) in serum were effectively reduced when DADS concentrations were 250,500,750 μmol/m L,and virus copy number was reduced. Therefore,three concentrations of DADS could effectively inhibit liver tumors development caused by Marek's disease virus in the early stage and had antiviral effect in vivo.
