摘要
目的:探讨阳和汤含药血清对乳腺癌干细胞HMLER90~(hi)增殖的影响及其作用机制。方法:将20只雌性SD大鼠随机分为阳和汤低、中、高剂量组和空白对照组,按低、中、高剂量分别灌胃阳和汤,空白对照组给予生理盐水,连续给药3天,制备阳和汤含药血清和空白血清。MTT法检测各组在含药血清干预24小时、48小时、72小时后对HMLER90~(hi)细胞增殖的影响;qRT-PCR检测各组细胞CD90、IL-8mRNA表达情况。结果:阳和汤含药血清干预24小时、48小时、72小时后,与空白对照组相比,阳和汤各剂量含药血清组细胞增殖的抑制率均增高,且对细胞增殖的抑制作用呈时间与剂量依赖性,差异具有统计学意义(P<0.05)。阳和汤各剂量含药血清组CD90、IL-8mRNA表达量均降低,且呈时间与剂量依赖性,差异具有统计学意义(P<0.05)。结论:阳和汤可抑制乳腺癌干细胞HMLER90~(hi)的增殖,其作用机制可能与其抑制单核巨噬细胞近分泌通路的传导有关。
        Objective: To investigate the effect of Yanghe Decoction serum on the proliferation of breast cancer stem cells HMLER90 hi and its mechanism. Methods: Twenty female Sprague-Dawley rats were randomly divided into low,medium and high dose groups of Yanghe Decoction and controlled group. The low-dose,medium-dose,and the high-dose group were for gavage with Yanghe Decoction,and the controlled group were given saline,continuous administration for 3 days. Preparation of Yanghe Decoction serum and blank serum. The effects of each group on the proliferation of HMLER90 hi cells at 24 h,48 h,and 72 h were detected by MTT assay. The expression levels of CD90 and IL-8 mRNA in each group was detected by real-time fluorescence quantitative PCR. Results: Compared with the controlled group after 24 h,48 h and 72 h of drug intervention,the inhibition rate of cell proliferation was increased in each dose group of Yanghe Decoction,and the inhibition of cell proliferation was time-and dose-dependent,and the difference was statistically significant(P<0.05). Compared with the controlled group,the expression levels of CD90 and IL-8 mRNA in each dose group of Yanghe Decoction decreased,and the difference was statistically significant(P<0.05). Conclusions: Yanghe Decoction can inhibit the proliferation of breast cancer stem cell HMLER90 hi,and its mechanism may be related to its inhibition of the conduction of the juxtacrine pathway of monocyte macrophage.
