摘要
目的分析人间充质干细胞成脂分化调控机制。方法传统方法培养人间充质干细胞,利用成脂诱导培养基诱导成脂分化,进行基因表达鉴定。结果实验组随着诱导时间的延长,脂滴逐渐显现、含脂滴的细胞数量明显增加,而未加成脂诱导剂的对照组细胞形态始终没有明显变化。实验组第7天、10天、14天的LPL、PPAR-γ基因表达高于对照组(无表达),同时随着时间的延长,表达信号强度明显逐渐上升,提示间充质干细胞成脂化与LPL、PPAR-γ基因表达有关。结论人间充质干细胞成脂分化可能与LPL、PPAR-γ基因表达增强有关。
        Objective To analyze the mechanism of adipogenic differentiation of human mesenchymal stem cells. Methods Human mesenchymal stem cells(HUMSCs) were cultured in the traditional method, adipogenic differentiation was induced by adipogenic medium, and gene expression was identified. Results With the prolongation of induction time, the lipid droplets gradually appeared in the experimental group, and the number of cells containing lipid droplets increased significantly. However, the cell morphology of the control group without addition of the lipidinducing agent remained unchanged. The expression of LPL and PPAR-γ on the 7 th, 10 th, and 14 th day in the experimental group was higher than that in the control group(no expression). At the same time, the intensity of the expression signal was gradually increased with time, suggesting the adipogenesis of mesenchymal stem cells and LPL, PPAR-γ gene expression is involved. Conclusion Adipogenic differentiation of human mesenchymal stem cells may be related to enhanced expression of LPL and PPAR-γ genes.
