摘要
为石榴良种的推广栽植和分子标记辅助选择育种奠定理论基础,采用ISSR分子技术方法,从ISSR随机引物中筛选出8条多态性好、谱带清晰、扩增稳定的引物(UBC834、UBC841、UBC811、UBC835、UBC827、UBC861、UBC840、UBC810)对秋艳、青丽和桔艳3个石榴良种进行ISSR-PCR扩增分析鉴别。结果表明:秋艳良种获得的2条特异性分子标记引物为UBC811和UBC834;青丽、桔艳良种获得的2条特异性分子标记引物为UBC835和UBC840,同时获得了3个石榴良种各自的特征图谱,初步建立了分子鉴别体系。
        The molecular identification of three good Punica granatum varieties(Qiuyan,Qingli and Juyan)were analyzed by using 8 primers(UBC834,UBC841,UBC811,UBC835,UBC827,UBC861,UBC840 and UBC810)with the advantages of good polymorphism,distinct band and stable amplification selected from ISSR random primers to lay the theoretical foundation for popularization,cultivation and molecular marker-assisted selection of good P.granatumvarieties.Result:Two specific molecular marker primers obtained from Qiuyan variety are UBC811 and UBC834.Two specific molecular marker primers obtained from Qingli and Juyan variety are UBC835 and UBC840.The specific chromatograms of three different P.granatum varieties are obtained. The molecular identification system is established preliminarily.
