摘要
将聚乙烯醇(PVA)和海藻酸钠(SA)混合作为细胞固定化载体,采用冷冻解冻法将喹啉降解菌(Achromobacter sp.)进行活细胞固定化,并对细胞固定化载体性能及喹啉降解特性进行了分析。结果表明:细胞固定化载体内部具有大量孔隙,比表面积达1.158 m~2/g;固定化细胞的力学强度和传质性能研究结果显示,经24 h高速搅拌,完整固定化细胞载体的保留率在98%以上,常温下对亚甲基蓝吸附的百分比为62%;当喹啉浓度小于500mg/L时,固定化细胞在16 h内对喹啉的降解率稳定在90%以上,且重复利用30次后,降解率仍能达到80%;与游离细菌相比,固定化细胞具有更强的适应环境及抗击负荷能力。
Polyvinyl alcohol(PVA)and sodium alginate(SA)were mixed as the cell immobilization carrier,and the quinoline-degrading bacteria(Achromobacter sp.)were immobilized by freezing-thawing method.The degradation characteristic of quinoline and performance of immobilized carrier were analyzed.The results showed that,The internal of cell immobilized carrier had a large number of pore structures,its specific surface area reached1.158 m~2/g.The study on the mechanical strength and mass transfer performance of immobilized carrier displayed that,after 24 hours of high-speed stirring,the retention rate of intact immobilized carrier was above 98%,and the percentage of methylene blue adsorption at room temperature was 62%.When the concentration of quinolinewas less than 500 mg/L,the degradation rate of quinoline in immobilized cells was more than 90%in 16 hours.Repeated used of 30 times,the degradation rate of quinolinecould still reach 80%,compared with free bacteria,immobilized cells had a stronger environment adaptation and resistance to load.
