摘要
目的:探讨miR-29、miR-200a在妊娠期糖尿病(GDM)患者外周血中的表达特征及临床意义。方法:选取本院接诊的80例GDM患者与80例糖耐量正常孕妇分别作为观察组与对照组,对两组孕妇进行外周血miR-29、miR-200a、miR-375检测,并进行血浆FINS、FPG、2hPG、HbAlc检测。分析miR-29、miR-200a、miR-375与血糖相关指标的相关性。结果:观察组FINS水平低于对照组,FPG、2hPG、HbAlc水平高于对照组(P<0.05)。观察组外周血miR-29表达量低于对照组,miR-375、miR-200a表达量均高于对照组(P<0.05)。相关性分析显示,miR-29表达与FPG、2hPG、HbAlc均呈正相关,与FINS呈负相关(P<0.05);miR-375、miR-200a表达均与FPG、2hPG、HbAlc呈负相关,与FINS呈正相关(P<0.05)。结论:外周血中miR-29、miR-200a、miR-375表达与血糖相关指标密切相关,miR-29、miR-200a、miR-375表达异常预示着GDM易感性,临床可通过测定外周血miR-29、miR-375表达为临床早期诊断GDM提供一定的参考依据。
        Objective:To investigate the expression features of miR-29 and miR-200 ain peripheral blood of pregnant women with gestational diabetes mellitus(GDM),and to study its clinical significance.Methods:80 pregnant women with GDM were selected into observation group,and another 80 normal pregnant women were included in control group.The expression of miR-29,miR-200 a,and miR-375 in peripheral blood of all included women were detected,and plasma FINS,FPG,2 hPG,and HbAlc levels of women in both groups were also tested.The expression of miR-29,miR-200 a,and miR-375,and the levels of plasma FINS,FPG,2 hPG,and HbAlc of women were compared between the two groups.The correlations between the expression of miR-29,miR-200 a,miR-375 and blood glucose related indicators were analyzed.Results:The level of FINS of women in observation group was significant lower than that of women in control group,but the levels of FPG,2 hPG and HbAlc of women in observation group were significant higher than those of women in control group(P<0.05).The expression of miR-29 in the peripheral blood of the women in the observation group was significant lower than that in the control group.The expression of miR-29 in the peripheral blood of women in observation group was significant higher than that of women in control group,but the expressions of miR-375 and miR-200 ain the peripheral blood of women in observation group were significant higher than those of women in control group(P<0.05).Correlation analysis showed that theΔCt value of miR-29 expression was positively correlated with level of FPG,2 hPG or HbAlc,but negatively correlated with level of FINS(P<0.05).TheΔCt values of miR-375 and miR-200 awere negatively correlated with level of FPG,2 hPG,or HbAlc,but positively correlated with level of FINS(P<0.05).Conclusion:The expression of miR-29,miR-200 aor miR-375 in peripheral blood of women with GDM is closely related to blood glucose related indicators.The abnormal expressions of miR-29,miR-200 aand miR-375 indicate the susceptibility of GDM.In clinical practice,the detection of the expression of peripheral blood miR-29 and miR-375 can provid a certain evidence for clinical diagnosis GDM early.
