摘要
目的探讨雷公藤甲素对肿瘤坏死因子-α(TNF-α)介导的HCM心肌细胞生长损伤的保护作用。方法采用四唑盐(MTT)比色法检测TNF-α抑制HCM细胞生长的半数抑制浓度(IC50)及雷公藤甲素干预对HCM细胞生长抑制率的影响;采用流式细胞术检测TNF-α联合雷公藤甲素对HCM细胞凋亡率的影响;使用相应试剂盒检测细胞上清液中肌酸激酶(CK)、乳酸脱氢酶(LDH)水平。结果 TNF-α对HCM细胞具有明显的生长抑制和凋亡诱导作用,且血清CK和LDH表达明显升高;低质量浓度雷公藤甲素干预后,TNF-α对HCM细胞的生长抑制和诱导凋亡作用均减弱(P <0. 01),血清CK和LDH表达呈下降趋势。结论低质量浓度雷公藤甲素可减弱TNF-α对HCM细胞的生长抑制和凋亡诱导作用。
        Objective To investigate the protective effect of triptolide on TNF-α-mediated damage in the growth of HCM cardiomyocytes.Methods MTT assay was used to detect the IC50 concentration of TNF-α inhibiting the growth of HCM cells, and the effect oftriptolide on inhibition rate of HCM cell growth,the flow cytometry was used to detect the effect of TNF-α combined with triptolide onthe apoptotic rate of HCM cells,and the corresponding kits were used to detect the levels of creatine kinase( CK) and lactate dehydro-genase( LDH) in cell supernatants. Results TNF-α had significant inhibition effect on the growth of HCM cells and significant induc-tion effect on the apoptosis of HCM cells,and the expression of serum CK and LDH was significantly increased. After intervention withlow concentrations of triptolide, the effect of TNF-α on the growth inhibition and apoptosis induction of HCM cells were bothweakened( P < 0. 01),the expression of serum CK and LDH showed a decreasing trend. Conclusion Low concentrations of triptolidecan attenuate the effect of TNF-α on inhibition of growth and apoptosis induction in HCM cells.
