摘要
目的:探讨蠲哮汤对哮喘大鼠半胱氨酸白三烯(CysLTs)和白细胞介素4(IL-4)、干扰素γ(IFN-γ)的影响及相关性。方法:将189只SPF级雌性SD大鼠随机分为正常组(A组),模型组(B组),孟鲁司特钠组(C组),地塞米松组(D组),蠲哮汤低、中、高剂量组(E、F、G组),卵蛋白氢氧化铝溶液致敏,并长期雾化吸入卵蛋白激发哮喘,A组予0.9%氯化钠溶液雾化代替,分别持续1、2、4周,各周结束时,处死每组大鼠9只,检测肺泡灌洗液(BALF)中CysLTs及IFN-γ、IL-4含量,并作相关性分析。结果:CysLTs:与A组同期比较,第1、4周各药物干预组降低,第2周各组升高(P﹤0.05);与B组同期比较,A组、蠲哮汤各剂量组均显著降低(P﹤0.05),各浓度组组间比较无显著性差异;D组低于其他药物干预组(P﹤0.05)。IL-4:与A组同期比较,第1、4周各药物干预组降低,第2周各组升高(P﹤0.05);与B组同期比较,A组、G组、D组均显著降低(P﹤0.05),各浓度组组间比较无显著性差异。CysLTs与IL-4呈正相关性、与IFN-γ呈负相关性。结论:IL-4、IFN-γ与CysLT通路相关,蠲哮汤可能是通过影响CysLTs干预哮喘炎症白三烯通道及通过影响IFN-γ、IL-4干预Th1/Th2失衡而达到治疗哮喘的目的。
        Objective: To investigate the effects and correlation of Juanxiao Decoction on CysLTs and IL-4, IFN-γ in asthmatic rats. Methods: A total of 189 SPF female SD rats were randomly divided into normal group(A group), model group(group B), montelukast group(group C), dexamethasone group(D group), low dose of Juanxiao Decoction group(group E), medium dose of Juanziao Decoction(F group) and high dose of Juanziao Decoction(G group), asthma model was established by ovalbumin sensitized aluminum hydroxide solution, and long-term inhalation of ovalbumin, Group A received 0.9% sodium chloride solution atomization respectively for 1, 2 and 4 weeks, at the end of each week, 9 rats in each group were sacrificed and the broncho alveolar lavage fluid(BALF) CysLTs and IFN-gamma, IL-4 content were detected as well as correlation analysis. Results: CysLTs: Compared with group A at the same time, the drug intervention group decreased at the 1 st and 4 th week, and increased at the 2 nd week(P<0.05). Compared with group B at the same time, group A and JuanXiao Tablets dosage groups were significantly lower(P<0.05), there was no significant difference between the concentration groups; group D was lower than other drug intervention groups(P<0.05). IL-4: Compared with group A at the same time, the levels of each drug intervention group decreased at the 1 st and 4 th week, and increased at the 2 nd week(P<0.05). Compared with group B at the same time, the levels of group A, group G and group D decreased significantly(P<0.05), and there was no significant difference among the groups with different concentrations. CysLTs was positively correlated with IL-4 and negatively correlated with IFN-γ. Conclusion: IL-4, IFN-γ and CysLTs channel may be interrelated, Juanxiao Decoction may through the intervention of CysLTs channel and leukotriene inflammation of asthma through the influence of IFN-γ, IL-4 and Th1/Th2 imbalance to treat asthma.
