摘要
目的探讨磷酸丝氨酸转氨酶1(phosphoserine aminotransferase 1,PSAT1)在非小细胞肺癌组织中的表达水平,并分析PSAT1表达与非小细胞肺癌预后的相关性。方法选择2013年12月—2015年10月确诊的非小细胞肺癌患者89例为观察组,同期25例肺良性病变患者为对照组。分别采用实时荧光定量PCR和免疫组织化学法检测和比较两组PSAT1 mRNA表达水平和蛋白阳性表达率;并分析PSAT1表达与患者病理特征的关系。结果观察组PSAT1 mRNA和蛋白阳性表达率显著高于对照组(P<0.05)。PSAT1蛋白表达与肿瘤分化程度、TNM分期以及是否淋巴结转移有相关性(P<0.05);而与患者的性别、年龄和肿瘤大小无相关性(P>0.05)。结论 PSAT1在非小细胞肺癌组织中呈过表达状态,检测PSAT1的水平对预测非小细胞肺癌进程及患者预后有重要意义。
        Objective To investigate expression level of phosphoserine aminotransferase 1( PSAT1) in nonsmall-cell lung cancer( NSCLC),and to analyze the relationship between PSAT1 protein and prognosis in NSCLC patients. Methods A total of 89 NSCLC patients during December 2013 and October 2015 were selected as observation group,and 25 patients with benign lung lesion at the same period were chosen as control group. Real-time fluorescent quantitation polymerase chain reaction( PCR) and immunohistochemistry method were respectively used to detect and compare expression level of PSAT1 mRNA and positive expression rate in two groups. The relationship between PSAT1 expression and clinicopathological characteristics was analyzed. Results Expression level of PSAT1 mRNA and positive expression rate in observation group were significantly higher than those in control group( P < 0. 05). Expression level of PSAT1 was correlated with differentiation degree,tumour node metastases( TNM) stage and lymph node metastasis( P <0. 05),but it was not correlated with gender,age or tumor size( P > 0. 05). Conclusion PSAT1 shows overexpression in NSCLC tissues. Detection of PSAT1 level is important for predicting NSCLC development and prognosis of patients.
