摘要
目的探讨转录因子GATA结合蛋白-3(GATA binding protein-3,GATA-3)在慢性鼻窦炎患者鼻黏膜中的表达及与白细胞介素(interleukin,IL)-5、IL-6、IL-8的关系。方法慢性鼻窦炎患者41例为观察组,同期行鼻内镜修补手术脑脊液鼻漏患者23例为对照组,采用免疫组织化学法检测2组鼻黏膜组织GATA-3蛋白阳性表达,实时荧光定量PCR法检测鼻黏膜组织GATA-3mRNA相对表达量,ELISA法检测血清IL-5、IL-6、IL-8水平;Pearson相关法分析2组鼻黏膜组织GATA-3蛋白阳性表达与血清IL-5、IL-6、IL-8的相关性。结果观察组鼻黏膜组织GATA-3蛋白阳性表达率(87.80%)、GATA-3mRNA相对表达量(0.60±0.12)及血清IL-5[(78.28±44.22)ng/L]、IL-6[(42.51±5.42)ng/L]、IL-8[(77.15±43.63)ng/L]水平均高于对照组[26.09%、0.13±0.11、(35.73±32.18)ng/L、(18.32±2.51)ng/L、(28.24±31.27)ng/L](P<0.05);观察组和对照组鼻黏膜组织GATA-3蛋白阳性表达与血清IL-5(r=0.761,P=0.002;r=0.814,P=0.001)呈正相关,与IL-6(r=0.021,P=0.518;r=0.019,P=0.594)、IL-8(r=0.061,P=0.411;r=0.072,P=0.395)无线性相关。结论慢性鼻窦炎患者鼻黏膜组织GATA-3表达上调,且与血清IL-5水平呈正相关,二者共同促进病情进展。
Objective To investigate the expression of transcription factor GATA binding protein-3(GATA-3)in the nasal mucosa in patients with chronic sinusitis and its relationship with the expression of serum interleukin(IL)-5,IL-6 and IL-8.Methods The positive expression of GATA-3 protein and relative expression of GATA-3 mRNA in nasal mucosa were detected by immunohistochemistry and real-time fluorescence quantitative PCR in 41 patients with chronic sinusitis(observation group)and 23 patients receiving nasal endoscopy for cerebrospinal rhinorrhea(control group).ELISA technique was used to detect the serum levels of IL-5,IL-6 and IL-8.The relationship of the positive expression of GATA-3 with IL-5,IL-6 and IL-8 was studied by Pearson correlation analysis.Results The positive expression rates of GATA-3 protein(87.80%),relative expression of GATA-3 mRNA(0.60±0.12),serum IL-5((78.28±44.22)ng/L),IL-6((42.51±5.42)ng/L)and IL-8((77.15±43.63)ng/L)in observation group were significantly higher than those in control group(26.09%,0.13±0.11,(35.73±32.18)ng/L,(18.32±2.51)ng/L,(28.24±31)ng/L)(P<0.05).The relative expression of GATA-3 protein in the nasal mucosa was positively correlated with serum IL-5(r=0.761,P=0.002;r=0.814,P=0.001),and was not linearly correlated with IL-6(r=0.021,P=0.518;r=0.019,P=0.594)and IL-8(r=0.061,P=0.411;r=0.072,P=0.395).Conclusion The expression of GATA-3 increases and is correlated with the level of IL-5,both of which are involved in the development of chronic sinusitis.
