摘要
目的探讨MDR1基因、CD269基因在老年骨髓瘤患者中的表达及与疾病预后的相关性。方法采用定量PCR法对2012-2013年我院收治的老年骨髓瘤患者体内MDR1基因与CD269基因表达进行检查,并分析表达结果与临床病理特征及预后的关系。结果 34例老年骨髓瘤患者血清及骨髓液样本中MDR1基因与CD269基因表达明显高于健康者。不同年龄、性别患者体内中MDR1基因与CD269基因表达差异无统计学意义,而出现肿瘤复发、Charlson指数(CCI)评分较高、生存时间较短的患者体内MDR1基因与CD269基因表达差异具有统计学意义(P <0.05)。其中MDR1基因与CD269基因表达越高的患者CCI评分≥5、生存时间越短、越易出现肿瘤复发。MDR1基因与CD269基因表达量与CCI评分呈正相关(r=0.786,P <0.001;r=0.825,P <0.001)。结论 MDR1基因、CD269基因参与骨髓瘤细胞的增殖与存活,影响骨髓瘤疾病预后,有望成为骨髓瘤未来诊断以及治疗的潜在靶标。
        Objective To investigate the expressions of MDR1 gene and CD269 gene in elderly myelomapatients and their correlation with disease prognosis. Methods RT-PCR was used to determine the expressions ofMDR1 and CD269 in elderly myeloma patients in hospital from 2012 to 2013.The associations between the expres-sions of MDR1 and CD269 in elderly myeloma patients and clinical and pathological features and prognosis wereanalyzed. Results The expressions of MDR1 gene and CD269 gene in serum and bone marrow samples of 34 elderlypatients with myeloma were significantly higher than those of healthy people. There were no significant differencesin the expressions of MDR1 gene and CD269 gene between different ages and genders. Patients with higher expres-sions of MDR1 gene and CD269 gene have higher tumor recurrence,with Clarson index(CCI)score more than 5 and shorter survival time(P < 0.05,respectively). The expressions of MDR1 and CD269 were positively correlatedwith CCI score(r = 0.786,P < 0.01;r = 0.825,P < 0.01). Conclusion MDR1 gene and CD269 gene are involvedin the proliferation and survival of myeloma cells,affecting the prognosis of myeloma diseases,which are expectedto be potential targets for future diagnosis and treatment of myeloma.
