响应面法优化南美白对虾头部多酚氧化酶的提取条件

摘要
[目的]运用响应面方法优化南美白对虾头部多酚氧化酶的提取条件。[方法]利用单因素试验和响应面优化法对浸提时间(t)、料液比、固体硫酸铵饱和度(%)等工艺条件进行分析与优化。[结果]南美白对虾头部多酚氧化酶提优化工艺参数:浸提时间66.61 min、料液比1:5.05(m/V)、固体硫酸铵饱和度48.79%,在此条件下获得酶比活力为98.87 U/mg,所得多酚氧化酶提取回归模型显著(R~2=0.9345),拟合性好,可用于预测多酚氧化酶的提取效果。
[Objective]The optimum conditions for the extraction of crude polyphenoioxidase(PPO) from Litopenaeus vannamei were determined using response surface methodology(RSM).[Methods]A box behnken design(BBD) was used to investigate the effects of three independent variables,namely extraction time(t),the weight ratio of cephalothoraxe-to-PBS(R) and solid ammonium sulfate saturation(%) on the response,PPO activities.The independent variables were coded at three levels and their actual values selected on the basis of preliminary experimental results.The BBD consisted of 17 experimental points and four replications at the center point.[Results]Data were analyzed using design expert and statistical analysis system software.A second-order polynomial model was used for predicting the response.Regression analysis showed that more than 93.45%of the variation was explained by the models.The optimal conditions for the PPO obtained using ridge analysis were 66.61 min,1:5.05,and 48.79%for Litopenaeus vannamei.Under the optimum conditions the corresponding predicted response values for PPO were 98.87U/min/ml.
