超低温冷冻保存对七带石斑鱼胚胎成活率的影响

本文利用6种小分子抗冻剂和4种大分子抗冻剂,通过单一、两种和三种抗冻剂组合使用,以及大分子与小分子抗冻剂混合使用,检测不同组合抗冻剂对七带石斑鱼胚胎孵化率的影响,从而筛选出适宜胚胎冷冻保存的玻璃液。结果表明;PM(24%PG+16%Me OH):Gly=3:1或4:1组成的玻璃化液中胚胎的孵化率较高。进一步实验显示PMG3S(35%PMG3+5%蔗糖)和PMG3T(35%PMG3+5%海藻糖)中胚胎成活率分别达可29.24±10.81%和27.01±3.39%。当利用PMG3S和PMG3T采用五步法分别处理肌节期和尾芽期胚胎时,当处理6min时胚胎体积脱水变小,之后逐渐恢复到正常体积,说明玻璃化液渗透进行胚胎细胞。利用PMG3S和PMG3T两种玻璃化液分别冷冻保存胚体形成期、肌节期和尾芽期胚胎,获得了82粒上浮胚胎,其中14粒胚胎继续发育,8粒尾芽期胚胎发育到心跳期,4粒胚体形成期胚胎发育到肌节期,2粒肌节期胚胎孵化出膜。
The effects of cryopreservation and the vitrification solution on the embryo hatchability of the seven-band grouper Epinephelusseptemfasciatus were evaluated in this study. Six small molecule cryoprotectants(PG, Me OH, Gly, DMF, DMSO and EG) and four macromolecularcryoprotectants(glucose, fructose, sucrose and trehalose) were used to determine the embryo toxicity levels. Results showed that the embryo survival rate was higher when the PM(24% PG + 16% Me OH):Gly ratios were 3:1 and 4:1. Further experiments showed that the embryo survival rates in PMG3S(35% PMG3 +5% sucrose) and PMG3T(35% PMG3 +5%trehalose) were relatively higher, which are 29.24±10.81% and 27.01±3.39%, respectively.
