人工培养多毛孢菌菌粉水提物对庆大霉素诱导LLCPK细胞凋亡及bFGFmRNA表达的影响

摘要
目的 研究人工培养的多毛孢菌菌粉水提物对庆大霉素诱导的猪肾小管上皮细胞(LLCPK)损伤、凋亡的保护作用及对碱性成纤维细胞生长因子(bFGF)mRNA表达的影响。
     方法 将培养的LLCPK细胞分成正常对照组、模型组和治疗组,模型组用庆大霉素刺激,治疗组同时加多毛孢菌菌粉水提物干预。①以MTT法观测存活细胞数;②并测定细胞上清液NAG酶浓度;③荧光标记细胞表面Annexin V,以细胞流式法测定其表达;④RT-PCR检测细胞bFGF mRNA的表达。
     结果 庆大霉素可引起细胞存活率的显著降低,NAG酶浓度显著增高,并提高细胞Annexin V的表达,和诱导LLCPK细胞bFGF mRNA的表达。治疗组经用多毛孢菌菌粉水提物干预后,则能提高肾小管上皮细胞LLCPK存活率,降低细胞凋亡数,并显著抑制bFGF mRNA的表达。
    
     结论多毛抱菌菌粉水提物能够对庆大霉素诱导
    的猪肾小管上皮细胞(LLCPK)损伤、凋亡起到保护作
    用,显著抑制细胞bFGF mRNA的表达可能与抗间质纤
    维化有关。
Objective To investigate the theraperatic effects of Cordyceps Sinensis in Gentamincin-induced cytotoxicity, apoptosis and bFGF gene expression in LLCPK.
    Methods Model group of LLCPK cells was stimulated with Gentamincin, and at same time added Cordyceps Sinensis in the theraperatic group. The number of cell survival was measured by MTT method and NAG level in culture supernatant was detected by color reaction. Flow cytometry was used to detect the Annexin V upon the cytomembrane. The bFGF mRNA expression was quantitated by RT-PCR assay.
    Results In this study, after treatment with Gentamincin, the number of cell survival was markly decreased, and NAG level was markly increased. Results presented that Gentamincin could induce the apoptosis and high expression of bFGF mRNA in LLCPK cells. And following incubation with Cordyceps Sinensis, the number of cell survival was significant increased, the level NAG and apoptosis and bFGF mRNA expression was decreased obviously . Cordyceps Sinensis could decrease the cell damage reduced by Gentamincin and inhibite the apoptosis and bFGF mRNA expression in LLCPK cells.
    Conclusion Cordyceps Sinensis can Proteel the cell damage,olecrease the cell's apoptosis and inhibite the bFGF mRNA expression in LLCPK cells induced by Gentamincin,that could be
    
    
    one of the mathanisms of its cutifibrosis.
