东北铁线莲地上部分化学成分研究

摘要
本文对东北铁线莲(Clematis manshurica Rupr.)地上部分(茎叶)的化学成分进行了研究,为扩大中药威灵仙的药物资源,变废为宝提供了科学依据。
     本文采用连续回流法提取东北铁线莲地上部分有效成分,用柱层析法,制备型薄层等方法对东北铁线莲地上部分有效成分进行分离纯化,并且采用高效液相色谱法、红外、紫外、质谱、核磁等光谱学手段鉴定了六个化合物的结构,分别是:β-谷甾醇(Ⅰ)、齐墩果酸(Ⅱ)、白头翁素(Ⅲ)、绿原酸(Ⅳ)、咖啡酸(Ⅴ)、阿魏酸(Ⅵ),其中化合物(Ⅳ)、(Ⅴ)、(Ⅵ)为首次在茎叶中发现;利用挥发油提取器和索氏提取器对其进行挥发性成分和脂肪油成分的提取,并对其进行了GC-MS的分析,鉴定出了24种挥发性成分和12种脂肪油成分;运用HPLC法测定出东北铁线莲地上部分中绿原酸的含量为1.36mg/g;用水提醇沉法提取两头尖多糖,运用紫外分光光度法对多糖含量进行测定,得到其多糖含量为1.6mg/g;通过ICP-MS对其含有的微量元素进行检测,检测结果为含有8种必需微量元素,其中铬(Cr)、锌(Zn)、铜(Cu)、镍(Ni)的相对含量较高;运用全自动氨基酸分析仪对氨基酸进行了初步研究,其含有16种必需氨基酸,总氨基酸含量为22.92ug/g。
     上述研究结果为东北铁线莲地上部分的开发利用提供了科学依据,为扩大威灵仙的药用资源奠定了基础,也使得地上部分能够变废为宝。
In this paper, the northeast wire Ephraim (Clematis manshurica Rupr.) theground part (stem) of chemical composition, which is expanding Chinese medicineWeiLingXian drug resources, reuse and offer scientific basis.
     In this article, the method of continuous reflux extraction clematis thenortheast part of Ephraim effective components, with column chromatography,preparation methods, such as type thin layer of the earth wire the northeastEphraim active ingredients of the purification, and by high performance liquidchromatography, infrared, ultraviolet, mass spectrometry, NMR spectroscopymeans such as identified six compound structure, it is respectively: betasitosterol (Ⅰ), oleanolic acid (Ⅱ), anemonin (Ⅲ), chlorogenic acid (Ⅳ),coffee acid (Ⅴ) and ferulic acid (Ⅵ), of compounds (Ⅳ),(Ⅴ),(Ⅵ) for thefirst time in the stem leaf found; Using naphtha and soxhlet extraction deviceis the volatile components and fat oil extraction of the ingredients, andanalyses the GC-MS analysis, identified the24kind of volatile organic compoundsand12kinds of fat oil component; Use contents were determined by HPLC methodfor the northeast part of the earth wire Ephraim the content of chlorogenic acidfor1.36mg/g; Water extraction and alcohol heavy law to extract pointy endspolysaccharide, using ultraviolet spectrophotometry were of polysaccharide, getthe contents of polysaccharide is1.6mg/g; Through the icp-ms to its containtrace elements for testing, test results to contain8kinds of necessary traceelements, including chromium (Cr), zinc (Zn), copper (Cu), nickel (Ni) therelative content of higher; Using automatic amino acid analyzer of amino acidwas studied, it contains16essential amino acids, total amino acids for22.92ug/g.
     The results of the study in the northeast part of the earth wire the effectiveuse of Ephraim provide the scientific basis for expanding WeiLingXian laid thefoundation of the medicinal resource, also make the ground part can reuse.
