中药肿节风药用成分的研究

摘要
肿节风是一味重要的传统中药,具有多种药理活性。本文通过DPPH法、脂质过氧化抑制试验、羟自由基清除试验测定了肿节风不同提取物的抗氧化作用,考察了它们的总多酚含量和总黄酮含量,采用MTT法体外抗肿瘤试验研究肿节风各个提取物的抗肿瘤效果。通过单因素试验和正交试验考察了肿节风乙醇提取工艺、超临界二氧化碳提取工艺的最优条件。并将最优工艺条件得到的提取物进行了抗氧化和抗肿瘤活性验证。通过GC-MS考察了肿节风超临界提取物和肿节风乙酸乙酯提取物的成分,并采用硅胶柱层析法对肿节风石油醚提取物进行了初步分离纯化。
     结果如下:
     (1)肿节风各提取物对DPPH自由基均显示了较好的清除作用,其中乙酸乙酯提取物的清除效果最佳;肿节风提取物显示了一定的脂质过氧化抑制作用,超临界提取物相对抑制脂质过氧化能力最好,在VC-Fe~(2+)的诱导下抑制率明显提高;肿节风各提取物显示了一定的羟自由基清除作用;肿节风乙酸乙酯提取物的总多酚和总黄酮含量最高;肿节风乙酸乙酯提取物有较好的体外抗HeLa、HepG-2肿瘤作用,肿节风乙酸乙酯提取物表现了较好的抗氧化和抗肿瘤活性,这可能和其所含的总黄酮含量有关,其内含的黄酮类物质是其抗氧化和抗肿瘤的主要药用成分之一。
     (2)肿节风总黄酮的最佳乙醇提取工艺条件是料液比1:35、乙醇浓度70%、提取温度80℃、提取时间2.5h。肿节风超临界二氧化碳萃取的最优工艺是以总黄酮、提取物质量为指标:萃取温度45℃,萃取器压力15MPa,乙醇夹带剂用量80ml,萃取时间1.5h;以异嗪皮啶的质量为指标:萃取温度35℃,萃取器压力25MPa,乙醇夹带剂用量60ml,萃取时间1h。肿节风乙醇提取最佳工艺条件制备得到的乙酸乙酯提取物经验证有较好的抗氧化、抗肿瘤效果。
     (3)气质联用分析肿节风超临界提取物和肿节风乙酸乙酯提取物,结果表明这两种提取物的成分有很大差异。石油醚提取物部分通过硅胶柱层析分离纯化得到一化合物,经鉴定为β-谷甾醇。
Sarcandra glabra (Thunb.) Nakai is an important crude herb used intraditional Chinese medicine, which has various pharmacological activiti-es. In this paper, the antioxidant ability of S. glabra extracts was assayedthrough 1, 1-diphenyl-2-picrylhydrazyl (DPPH) method, lipid peroxidationinhibiting test, hydroxyl free radical scavenging test. The total phenolic andtotal flavones contents of the extracts were assayed. The antitumor activityof the extracts was tested by MTT method. The optimum extractionprocesses of medicinal components in S. glabra by ethanol extraction andsupercritical carbon dioxide fluid extraction were chosen through singlefactor and orthogonal experiments. The antioxidant and antitumor activitiesof the extracts under the optimum extraction processes were validated.
     The results were showed as followed:
     (1) All S. glabra extracts showed good DPPH radical scavenging effects, while the ethyl acetate extract showed best effect. All S. glabra extractsshowed lipid peroxidation inhibiting ability in certain extent, while theextracts of supercritical carbon dioxide fluid extraction showed best effect.The inhibiting rate was enhanced under the inducement from Fe~(2+)-VCobviously. All S. glabra extracts showed hydroxyl free radical scavengingability in certain extent. The ethyl acetate extract had the top content of totalphenolic and total flavones, and it inhibited the growths of HeLa andHepG-2 cells. It indicated that antioxidant and antitumor activities of theextracts were related with total flavones content. Total flavones in theextracts of S. glabra was one of the main medicinal components in S.glabra.
     (2) The optimum ethanol extraction process of total flavones from S.glabra was adding 35 times amount of 70% ethanol and extracting for 2.5hours at 80℃. The optimum supercritical carbon dioxide fluid extractionprocess was 45℃, 15MPa, 80ml ethanol and 1.5 hours, evaluated by bothtotal flavones content and extract content. The optimum supercritical carbondioxide fluid extraction process was 35℃, 25MPa, 60ml ethanol and 1 hour,evaluated by isofraxidin content. The ethyl acetate extract gained from theoptimum ethanol extraction process was proved to have good antioxidantand antitumor activities.
     (3) The chemical compositions of the extract from supercritical carbondioxide fluid extraction and ethyl acetate extract were separated and identified by GC-MS. The results showed great difference between twoextracts. Petroleum etherp extract was isolated by repeatedly silica gelcolumn chromatography, and one compound was isolated and identified asβ-sitosterol.
