摘要
[目的]建立金钱草茎段组织培养再生体系。[方法]于4、5月取当年生金钱草茎段为外植体,MS作为基础培养基,研究不同植物生长调节剂对诱导、增殖、生根过程的影响。[结果]MS+6-BA 1.0 mg/L+IBA 0.20~0.50 mg/L适合金钱草不定芽诱导,诱导率为100%;MS+6-BA1.0 mg/L+IBA 0.10~0.20 mg/L适合继代增殖培养; 1/2MS+IBA0.50 mg/L+NAA0.1~0.5 mg/L适合不定根诱导,诱导率高达100%;组培苗移植到盛有腐殖质花盆中,成活率为100%。[结论]该研究可为金钱草野生种质资源保存和优质种苗生产提供技术参考。
[Objective]To establish the tissue culture regeneration system of Lysimachia christinae Hance.stem.[Method]In April and May,the stems of L. christinae were used as explants and MS as basic medium to study the effects of different plant growth regulators on induction,proliferation and rooting. [Result]MS+6-BA 1.0 mg/L+IBA 0.20-0.50 mg/L was suitable for adventitious bud induction of L. christinae,the induction rate was 100%. MS+6-BA 1.0 mg/L+IBA0.10-0.20 mg/L was suitable for seedling subculture. 1/2MS+IBA0.50 mg/L+NAA 0.1-0.5 mg/L was suitable for root induction culture,the induction rate was 100%. The tissue culture seedlings were transplanted to pots in flowerpot with humus and the survival rate was 100%. [Conclusion]The study can provide technical reference for the conservation of wild germplasm resources and the production of high-quality seedlings of L. christinae.
