摘要
以蓝莓试管苗为材料,筛选适合瓶内生根技术的方法、培养基及培养条件,为蓝莓组培快繁技术提供理论指导.通过在改良的WPM(木本植物培养基)中添加不同浓度的IBA(吲哚丁酸)、NAA(萘乙酸)、活性炭等研究蓝莓试管苗瓶内生根技术,采用正交实验设计,筛选最适蓝莓试管苗瓶内生根培养条件.结果表明:蓝莓试管苗在改良的1/2WPM培养基中生根率高,试管苗的上端和下端对蓝莓的生根率无明显影响,蓝莓生根最适培养基为改良1/2 WPM+IBA 0.8 mg/L+NAA 0.20 mg/L+活性炭0.06%+暗处理14 d,60 d后生根率达83.3%.
Blueberry test-tube seedling was cultured in different culture medium and conditions to explore the appropriate test-tube rooting method and provide theoretical basis for blueberry rapid propagation.IBA,NAA and activated carbon with different concentrations were added into the improved WPM(Woody plant culture medium)and then orthogonal experiment design determined the optimal rooting culture conditions of blueberry in vitro plantlets.The results showed that the rooting rate of blueberry test-tube plantlets was relatively higher in the improved 1/2 WPM medium and the rooting rate was not significantly affected by the upper and lower ends of the test-tube plantlets.The optimal rooting medium of blueberry was improved 1/2 WPM+IBA 0.8 mg/L+NAA 0.2 mg/L+activated carbon 0.06%+dark treatment for 14 days.In this condition,the rooting rate reached 83.3% after 60 days cultivation.
