参苓白术散对CD大鼠血清及结肠组织中IL-1β、IL-4及IL-10含量的影响

摘要
目的:通过观察参苓白术散对克罗恩氏病(CD)大鼠血清及组织中白介素1β、白细胞介素4(IL-4)及白细胞介素10(IL-10)含量的影响,探讨参苓白术散治疗克罗恩氏病的效应机制。方法:受试大鼠按随机数字表法分为空白组和模型组,采用三硝基苯磺酸(TNBS)合乙醇溶液灌肠制备克罗恩氏病大鼠模型。造模成功后将模型大鼠分为模型组、参苓白术散组和阳性对照组(美沙拉嗪组),空白组和模型组大鼠每日给予10ml/kg蒸馏水灌胃,参苓白术散组给予12g·kg-1·d-1参苓白术散药剂灌胃,阳性对照组给予0.2g·kg-1·d-1美沙拉嗪颗粒灌胃,持续治疗21d后,分别对各组大鼠血清及组织中IL-1β、IL-4和IL-10含量进行检测。结果:与空白组比较,模型组大鼠一般生存状况较差,血清及组织中IL-1β含量显著升高,IL-4和IL-10含量降低(P<0.05);与模型组比较,参苓白术散组大鼠一般生存状况明显改善,血清及结肠组织中IL-1β含量显著降低(P<0.05),而IL-4和IL-10含量升高(P<0.05)。结论:参苓白术散具有改善克罗恩氏病大鼠一般生存状况,调节结肠组织Th1/Th2细胞因子平衡分泌的作用。
        Objective: To study effects of Shenling Baizhu San(SLBZ) on IL-1β, IL-4 and IL-10 contents in colon serum and tissue of rats with Crohn's disease. Methods: Rats were randomly divided into 2 groups: normal group and model group. After UC modle was successful, UC rats were numbered and grouped by random number table into the model group, SLBZ group and positive group; then rats in normal and model group were intragastrically administrated with distilled water 10 ml/kg every day, SLBZ group with SLBZ volume of 12g·kg-1·d-1, and positive group with suspension of mesalazine 0.2g·kg-1·d-1, for 21 consecutive days. Levels of IL-1β, IL-4, and IL-10 in colon serum and tissue of rats in each group were detected. Results: In successful model group, generally living conditions of rats were poor, levels of IL-1β in colon serum and tissue were much higher(P<0.01); but levels of IL-4 and IL-10 were lower than those in the normal group(P<0.05). Compared with the model group, the general survival of the state in the SLBZ San group was improved significantly. SLBZ could increase levels of IL-4 and IL-10 significantly(P<0.05), decrease levels of IL-1β(P<0.05). Conclusion: It was suggested that SLBZ has functions of improving general living conditions, adjusting cytokine balance of TH1/TH2.
