常春藤皂苷元(hederagenin)通过抑制STAT3通路降低CaSki宫颈癌细胞增殖能力并促进其凋亡

摘要
目的探讨常春藤皂苷元(hederagenin)对宫颈癌细胞活性及凋亡的影响及机制。方法培养CaSki宫颈癌细胞,采用(0、 10、 20、 40、 80)μg/mL常春藤皂苷元处理,噻唑蓝(MTT)法测定细胞活性,确定其半数抑制剂量约40μg/mL。采用40μg/mL常春藤皂苷元处理宫颈癌细胞,流式细胞术测定宫颈癌细胞凋亡、 Western blot法测定宫颈癌细胞裂解型胱天蛋白酶3(c-caspase-3)、 c-caspase-9、信号转导子和转录激活因子3(STAT3)、磷酸化的STAT3(p-STAT3)蛋白水平。用STAT3信号通路抑制剂AG490和常春藤皂苷元共同处理宫颈癌细胞,用上述方法检测细胞活性和凋亡水平。结果与对照组细胞相比, 40μg/mL常春藤皂苷元处理后的宫颈癌细胞增殖活性降低,细胞凋亡水平升高, c-caspase-3和c-caspase-9水平增加, p-STAT3蛋白水平降低。与常春藤皂苷元处理细胞相比, AG490和常春藤皂苷元共同处理的宫颈癌细胞活性进一步降低,细胞凋亡率增加,细胞中c-caspase-3和c-caspase-9的蛋白水平升高。结论常春藤皂苷元抑制宫颈癌细胞的增殖并诱导其凋亡,与抑制STAT3信号通路有关。
        Objective To investigate the effect of hederagenin on the activity and apoptosis of cervical cancer cells and its mechanism. Methods Cervical cancer Ca Ski cells were cultured and treated with 0,10,20,40,80 μg/m L hederagenin.Cell viability was determined by MTT assay. The 50% inhibitory concentration( IC50) was determined to be about 40 μg/m L.The cervical cancer cells were treated with 40 μg/m L hederagenin. The apoptosis of cervical cancer cells was measured by flow cytometry,and the levels of cleaved caspase-3( c-caspase-3),c-caspase-9,and STAT3 and p-STAT3 proteins were determined by Western blot analysis. STAT3 signaling pathway inhibitor AG490 and hederagenin were used to treat cervical cancer cells,and then the activity and apoptotic level of cervical cancer cells were detected by the above methods. Results Compared with the control cells,the proliferation activity of the cervical cancer cells treated with 40 μg/m L hederagenin decreased,the apoptotic level increased,the levels of c-caspase-3 and c-caspase-9 increased,and the level of p-STAT3 protein decreased. Compared with the cells treated with hederagenin,the activity of cervical cancer cells treated with AG490 and hederagenin decreased further,the apoptotic rate increased,and the protein levels of c-caspase-3 and c-caspase-9 increased.Conclusion Hederagenin inhibits the proliferation and promotes the apoptosis of cervical cancer cells,which is related to the inhibition of STAT3 signaling pathway.
