四逆汤对心肌梗死后慢性心衰大鼠心功能和氧化应激反应的作用研究

摘要
目的观察四逆汤对心肌梗死后慢性心衰大鼠心功能和氧化应激反应的影响。方法采用结扎大鼠冠状动脉导致急性心肌梗死制备慢性心衰大鼠动物模型,将造模成功、存活的大鼠随机分为模型组、盐酸贝那普利组、四逆汤高剂量组、四逆汤中剂量组和四逆汤低剂量组,另设一组未行冠脉结扎术的大鼠为假手术组,每组8只。各组予以相应药物或去离子水,连续灌胃8周后行大鼠心脏彩色多普勒超声检查、病理形态学观察,并比较各组大鼠血清超氧化物歧化酶、丙二醛、谷胱甘肽过氧化物酶、及过氧化氢的含量差异。结果给药8周后,小动物心动超声结果显示:与假手术组比,模型组大鼠的心脏左室收缩末期内径显著升高(P<0.05),左室射血分数、左室短轴收缩率、舒张末期室间隔厚度、收缩末期室间隔厚度显著降低(P<0.05);与模型组比,四逆汤高、中剂量组左室射血分数、左室短轴收缩率、舒张末期室间隔厚度、收缩末期室间隔厚度显著改善(P<0.05),而低剂量四逆汤有改善左室射血分数、左室短轴及收缩率的趋势,但差异没有统计学意义(P>0.05)。HE染色示:给药组的大鼠心肌结构破坏程度较模型组明显改善;Masson染色示:四逆汤可有效降低心肌组织胶原蛋白含量,抑制心衰大鼠心肌纤维化(P<0.05)。结论四逆汤能够改善心梗所致的慢性心衰大鼠的心功能,其机制可能与改善氧化应激状态,提高抗氧化能力,减少氧化损伤相关。
        Objective To observe the effects of Chinese Sini decoction(SNT) on cardiac function and oxidative stress in rats with chronic heart failure after myocardial infarction. Methods Rat model of chronic heart failure and of acute myocardial infarction,caused by ligation of rat coronary artery,were prepared. The survivals were randomly divided into model group, Benner Pury group,SNT high dose group, SNT medium dose group and SNT low dose group. Each group has 10 rats.Rats with no coronary artery ligation was adpoted as Sham operation group. Each group were given relative medicine or deionized water. After 8 weeks of gavage,then rats were evaluated by heart color doppler echocardiography and observed by pathomorphology. Then, the differences of serum SOD、MDA、GSH-Px and H_2O_2 levels were compared. Results After 8 weeks of administration, the heart LVIDs of the rats showed that the model group significantly increased(P<0.05), compared with the sham operation group. LVEF%、LVFS%、IVSd and IVSs decreased significantly in the model group(P<0.05). Compared with the model group, LVEF%、LVFS%、IVSd、IVSs(P<0.05) could improve LVEF%、LVFS%、IVSd、IVSs(P<0.05) by SNT of high dose and medium dose. LVEF%、LVFS% trend could also be improved by low dose SNT. But the differences were not statistically significant(P >0.05). Compared with the model group, the myocardial structural damage in the HE staining administration group was improved. Masson staining showed that the SNT could effectively reduce the collagen content of myocardial tissue and inhibit myocardial fibrosis in rats with heart failure(P<0.05). Conclusion The SNT can improve the cardiac function in rats with chronic heart failure. The mechanism may be related to improving oxidative stress, enhancing antioxidant capacity and reducing oxidative damage.
