摘要
目的检测宫颈癌中SIX1和Ezrin的表达,探讨其在宫颈癌浸润转移中的作用及临床意义。方法采用免疫组化检测2015年10月至2017年10月秦皇岛市第一医院确诊的80例宫颈癌组织和配对癌旁组织中SIX1和Ezrin的表达。应用RNA干扰(RNAi)技术,将SIX1-si RNA表达质粒转染宫颈癌Hela细胞,采用RT-PCR和Western blot检测沉默SIX1基因后宫颈癌Hela细胞中SIX1和Ezrin的m RNA和蛋白表达水平。结果宫颈癌组织SIX1的阳性表达率高于癌旁组织,Ezrin的中-强度阳性表达率高于癌旁组织,差异有统计学意义(P <0.05);二者的表达与宫颈癌FIGO分期和淋巴结转移有关。转染SIX1-si RNA后宫颈癌Hela细胞SIX1和Ezrin的蛋白水平有显著下调。结论 SIX1可能通过调节其下游Ezrin的表达在宫颈癌的发生、发展过程中发挥重要作用。
        Objective To investigate the expression the clinical significance of SIX1 and Ezrin in cervical cancer.Methods Immunohistochemical(IHC) staining was used to detect the expressions of SIX1 and Ezrin in 80 cervical cancer tissues and 80 corresponding paracancer tissue collected from October 2015 to 2017.SIX1 siRNA was constructed by RNA interference and then stably transfected into Hela cells.The expression of SIX1 and Ezrin mRNA and protein was detected by RT-PCR and Western blot respectively.Results The positive rate of SIX1 and the moderate-strong positive rate of Ezrin in cervical cancer was signif icantly higher than that of paracancer tissues,the difference was statistically signif icant(P <0.05).SIX1 and Ezrin protein expression were related to FIGO stage and lymph node metastasis of cervical cancer.The Protein levels of SIX1 and Ezrin were down-regulated in Hela cells after RNAi of SIX1.Conclusin SIX1 may play an important role in the occurrence and development process of cervical cancer by regulating its downstream of Ezrin.
