摘要
目的应用超声造影技术定量分析L-精氨酸对大鼠肾缺血再灌注损伤肾皮质血流灌注的影响,探讨超声造影技术在评价药物干预肾缺血再灌注损伤疗效中的应用价值。方法健康雄性大鼠24只随机分成假手术组(Sham组)、缺血再灌损伤组(I/R组)和L-精氨酸组(L-arg组),每组8只。行超声造影检查观察肾皮质血流灌注情况并脱机分析时间-强度曲线(TIC)。实验结束后,行肾组织病理学检查。结果与假手术组比较,I/R组和L-arg组肾脏病理改变明显,达峰时间(TTP)和平均通过时间(MTT)延长,曲线下面积(AUC)显著增加(P<0.05);与I/R组比较,L-arg组病理改变显著,峰值浓度(Peak)增高,TTP、MTT缩短,AUC减少(P<0.05)。结论超声造影结合时间-强度曲线能定量分析L-精氨酸对大鼠肾缺血再灌注损伤肾皮质血流灌注的改善。
        Objective The application of contrast-enhanced ultrasound to quantitatively analyze the effect of L-arginine on renal cortex blood perfusion in rats with renal ischemia-reperfusion injury,and to evaluate the value of contrast-enhanced ultrasound in the evaluation of drug intervention in renal ischemia-reperfusion injury.Methods Twenty-four healthy male rats were randomly divided into sham-operated group(Sham group),ischemia-reperfusion injury group(I/R group)and L-arginine group(L-arg group),with 8 rats in each group.Contrast-enhanced ultrasound was performed to observe the renal cortex blood flow perfusion and the time-intensity curve(TIC)was analyzed off-line.At the end of the experiment,renal histopathological examinations were performed.Results Compared with the sham-operated group,the pathological changes of the kidney in the I/R group and the L-arg group were significant,the peak time(TTP)and mean transit time(MTT)were prolonged,and the area under the curve(AUC)was significantly increased(P<0.05);Compared with the I/R group,the pathological changes in the L-arg group were significant,Peak increased,TTP,MTT shortened,and AUC decreased(P<0.05).Conclusions Contrast-enhanced ultrasound combined with time-intensity curve can quantitatively analyze the improvement of renal cortical blood flow in rats with renal ischemia-reperfusion injury induced by L-arginine.
