当归对支气管哮喘模型小鼠的平喘作用及对糖皮质激素所致阴虚症状的缓解作用研究

摘要
目的研究当归对支气管哮喘模型小鼠的平喘作用及对糖皮质激素所致阴虚症状的缓解作用。方法将48只小鼠随机分为空白组、模型组、氢化可的松组、当归+氢化可的松组4组,每组12只,采用卵蛋白通过致敏、激发复制哮喘模型。从第15天开始,当归+氢化可的松组灌胃当归水煎液(8 g/kg),连续7 d;从第17天开始,氢化可的松组、当归+氢化可的松组灌胃氢化可的松琥珀酸钠混悬液(0.05 g/kg),连续5 d。空白组及模型组灌胃等量生理盐水。比较各组小鼠体质量、进食和饮水量、自主活动情况、肺组织病理学变化及血清环磷酸腺苷(c AMP)与环磷酸鸟苷(c GMP)水平。结果与空白组比较,模型组小鼠肺组织出现明显的病理变化,支气管壁充血、结构紊乱,上皮细胞肿胀,基底膜增厚、边缘模糊;氢化可的松及当归+氢化可的松均可使上述肺组织的病理变化得到一定程度的改善,且当归+氢化可的松的改善作用优于氢化可的松。与模型组比较,氢化可的松组小鼠体质量明显下降,进食和饮水量、站立次数、活动次数均明显增加,血清c AMP水平、c AMP/c GMP明显升高,2组比较差异均有统计学意义(P<0.05或P<0.01);在当归的干预下,当归+氢化可的松组小鼠的体质量明显升高,进食和饮水量、站立次数、活动次数明显减少,血清c AMP水平、c AMP/c GMP明显降低,与氢化可的松组比较差异均有统计学意义(P<0.05或P<0.01)。结论当归与糖皮质激素对支气管哮喘模型小鼠均具有一定的平喘作用,且当归可明显缓解糖皮质激素治疗支气管哮喘过程中所致的阴虚症状。
        Objective To observe the effect of Danggui( Radix Angelicae Sinensis)( RASI) on mice with bronchial asthma and its mitigative effect on glucocorticoid-induced yin deficiency symptoms during treatment.Methods 48 mice were randomly divided into the blank group,the model group,the hydrocortisone sodium succinate( HSS) group and the RASI+HSS group with 12 mice in each. These asthma model mice were replicated by the stimulation and sensitization of ovalbumin( OVA). On the 15 th day,mice in the RASI+HSS group were treated with8 g/kg RASI constantly for 7 days; on the 17 th day,the HSS group and RASI+HSS group were given 0.05 g/kg HSS constantly for 5 days; At the same time,the blank group and the model group were given the same amount of normal saline. Finally,animal body weight,food and water intake,autonomic activity,lung histopathological changes,and serum c AMP and c GMP were measured. Results Compared with the blank group,in the model group,some noticeable pathologic changes in the lung tissue of asthmatic mice were observed,like bronchial wall congestion,structural disorder,epithelial cell swelling,thickened basement membrane,and fuzzy edge. Under the intervention of HSS and RASI+HSS,these pathological changes have been alleviated,and the latter has taken effect better than the former.Compared with the asthma model group,the body weight of mice in the HSS group was significantly decreased,and the intake of food and drinking water,the number of autonomic activity and c AMP,c AMP/c GMP in serum increased significantly,differences had significance( P< 0.05 or P < 0.01). However,having given RASI,these changes have been clearly reduced with statistically significant differences( P < 0. 05 or P < 0. 01). Conclusion RASI and glucocorticoid have a certain antiasthmatic effect on mice with bronchial asthma,and RASI can relieve glucocorticoidinduced yin deficiency symptoms during treatment.
