摘要
优化Folin-Ciocalteu比色法测定石榴花提取物中多酚含量的条件,并对该方法进行系统评价。以没食子酸为标准物质,确定测定波长后系统考察Folin-Ciocalteu试剂(FC试剂)浓度、Na2CO3试剂浓度、反应时间、反应温度等因素对测定结果的影响,并对该方法的稳定性、精密度、回收率等进行试验。研究表明,石榴花提取物多酚含量测定的适宜条件为:1 m L样品液、5 m L 40%FC试剂、4 m L 10%Na_2CO_3试剂、25~℃避光反应100 min,测定波长为750 nm,该方法的稳定性、精密度、回收率均较好。
        The optimum conditions for the measurement of polyphenol in pomegranate flowers by Folin-Ciocalteu colorimetry were optimized,and the method was evaluated systematically. The effects of the concentration of FC reagent,concentration of Na2 CO3 reagent,reaction time and reaction temperature on the results were studied by using gallic acid as the reference material. The stability,precision and recovery rate of the method were tested. It showed that the optimum conditions for the measurement of polyphenols in pomegranate flower extracts were:1 m L sample solution,5 m L 40 % FC reagent,4 m L 10 % Na_2 CO_3 reagent,which were reacted at 25~℃ for 100 min,and the measurement wavelength was 750 nm. The stability,precision and recovery of the method are good.
