摘要
为了分析H1N1猪流感病毒(SIV)介导小鼠mmu-miRNA-206-3p(mmu-miR-206-3p)下调及其对纤连蛋白1(FN1)表达过程的影响,试验选用10只6周龄雌性SPF级Balb/c小鼠建立感染H1N1 SIV小鼠模型,并将小鼠分为对照组和攻毒组,对照组用无病毒的尿囊液滴鼻,攻毒组用病毒尿囊液滴鼻,观察并记录小鼠的表现,利用RT-qPCR、Western-blot方法检测mmu-miR-206-3p、FN1和纤连蛋白1基因(Fn1)的变化。结果表明:感染H1N1 SIV 48 h后,对照组小鼠无异常表现,攻毒组小鼠精神萎靡,扎堆,食欲减退,被毛逆立。与对照组相比,12 h后攻毒组mmu-miR-206-3p相对表达量下降, FN1相对表达量升高,Fn1相对表达量变化不显著。说明H1N1 SIV可抑制mmu-miR-206-3p表达,减少mmu-miR-206-3p对Fn1翻译的抑制,进而导致FN1的相对表达量升高。
        To analyze the down-regulation of mouse mmu-miRNA-206-3 p(mmu-miR-206-3 p) mediated by H1 N1 swine influenza virus and its effect on FN1 expression,ten 6-week-old female SPF-level Balb/c mice were used to establish a mouse model. The mice were divided into the control group and the challenge group. The control group used a virus-free allantoic droplet nose, and the challenge group used virus urine. The state of the mice was observed. The changes of mmu-miR-206-3 p, FN1 and Fn1, were detected by RT-qPCR and Western-blot. The results showed that there was no abnormality in the control group after 48 hours of infection with H1 N1 virus. The mice in the challenge group were apathetic, tied up, loss of appetite, and the coat was reversed. After 12 h, the expression level of mmu-miR-206-3 p was decreased and the expression of FN1 was increased in the challenge group. There was no significant difference in the expression of Fn1 gene in the challenge group compared with the control group. It indicates that H1 N1 swine influenza virus inhibits the expression of mmu-miR-206-3 p, which may reduce its inhibition of Fn1 translation, which leads to an increase in the expression of FN1.
