蛋氨酸螯合铬食品添加剂的研究

摘要
铬(Chromuim,Cr~(3+),后述铬均为三价铬)是人体必需的微量元素之一,是一系列酶和蛋白的活性中心的组成部分。对于维持机体三大物质代谢和正常生理状况起到重要作用。不仅具有增强清除体内自由基的作用,还有抑制自由基产生的作用。蛋氨酸(Metnionine,Met)是维持人体正常生理机能的必需氨基酸,具有一定的生物学活性,而且是铬的天然配位体和有效的自由基清除剂。蛋氨酸螯合铬把人体必需的蛋氨酸和微量元素铬(Cr~(3+))有机的结合起来,增强了二者的协同作用,因其在人体中特定的吸收与转运模式,从而成为一种新型高效的铬营养强化剂。
     本研究以蛋氨酸和六水合三氯化铬为原料,通过化学反应合成,经水洗醇提制备蛋氨酸铬的螯合物。正交试验优化得最优的反应条件为——Met:Cr的配位摩尔比是3:1,pH=7.0,反应温度80℃,反应时间为60 min,蛋氨酸浓度为15%,产率为48.41%。反应产物分离纯化后,利用氨基酸组成分析、元素分析、紫外光谱分析、红外光谱分析等手段初步确定产物为蛋氨酸螯合铬(配位比为3:1)。在此基础上经质谱分析推断其分子式为:C_(15)H_(30)N_3O_6S_3Cr;结构式为:[Cr(NH_2CHCH_2CH_2SCH_3COO)_3];相对分子质量:M=497.0。
     蛋氨酸螯合铬的熔点为:352~356℃。蛋氨酸整合铬在37℃时的溶解度为42mg/100mL。(按照食品添加剂的国标要求)利用动物试验对其安全性进行了评价,结果表明蛋氨酸铬是安全的。同时将其以不同浓度添加到奶粉中进行应用试验,通过测定奶粉中的一些营养指标(乳脂肪、V_E、V_(B2)、V_(B6)、V_(PP)),确定蛋氨酸铬螯合物对奶粉中V_(B2)、V_(B6)和V_E有破坏作用,而对V_(PP)、乳脂肪无破坏作用。如忽略对V_(B2)、V_(B6)和V_E的影响,可将蛋氨酸铬螯合物应用于糖尿病人专用奶粉中,这一课题将有着广阔的市场前景。
Chromium(Chromuim,Cr3+) was one of the microelements necessary for human body and the component of alive center of series of enzymes and protein. Chromium made a very important role in maintaining matter metabolism and physiological function in human body. It could not only eliminate free radical but also restrain the production of free radical. Methionine was an essential amino acid in human body, it possessed some biological activity and was natural Hgand of chromium. Methionine could also eliminate free radical effectively. Methionine was combined with chromium for their special absorption and transport in body, which strengthened their biological function. Chromium-Methionine chelate was a new nutritional intensifying agent.
    The Chromium-Methionine chelate was synthesized by methionine and chromium chloride. The final reactant was washed by water and ethanol, and then we got the Chromium-Methionine chelate.The optimum conditions were that molar ratio of methionine to chromium-chloride was 3:1, reaction temperature was 80癈, reaction pH was 7.0, reaction time was 60 min and the concentration of methionine was 15 percent. The yield was 48.41%. Composition and structure of the complex were characterized by amino acid analysis, elementary analysis, UV spectrogram analysis, infrared spectrum and mass spectrum analysis. Chromium-Methionine's molecule composition was [Cr(NH2CHCH2CH2SCH3COO)3 ] . Relatively molecule mass was 497.0.
    The melting point of Chromium-Methionine chelate was 352-356 .The solubility of Chromium-Methionine chelate was 42.00mg/100g H2O.
    The security of complex was appraised by animal experiment. The results made clear that Chromium-Methionine chelate was safety. Milk powder was added with different concentration of complex at the same time. By determining some nutrition index of milk powder such as fat, VE, VB2, VB6, VPp and so on, we could make sure the complex destroied VB2, VB6 and VE, but it didn't destroy VPP and fat. If we ignored effects on VB2, VB6 and VE, we could use Chromium-Methionine chelate as additive for milk power of diabetic.
