山羊FSHβ基因多态性及其与产羔数相关性分析

摘要
促卵泡素是一种垂体糖蛋白促性腺激素,影响动物的生殖系统发育和雄性动物精子的生成,常用于常用与胚胎移植和超数排卵,是促进卵泡的生长、发育、成熟和排卵必不可少的激素。天府肉羊是四川农业大学历经28年采用多品种复杂育成杂交育种技术选育成功的肉山羊新品系。本实验以四川农业大学羊业科教中心和汇源牧业有限公司培育的166只天府肉羊、101只波尔山羊和67只成都麻羊为研究对象,对FSHβ基因PCR-SSCP的遗传多样性进行研究及其与产子数的相关性分析。本研究参照GeneBank中的牛、绵羊和猪等物种的FSHβ基因序列,设计PCR引物,扩增山羊FSHβ基因的5′调控区、外显子1和外显子2。结果表明:
     FSHβ基因的5′调控区,发现1个SNP位点,是A712G。FSHβ基因引物1在天府肉羊和波尔山羊中均为中度度多态(0.25<PIC<0.5),在成都麻羊中为低度多态(PIC<0.25)。x 2适合性检验表明:引物1扩增片段均在波尔山羊中极显著的偏离了Hardy-Weinbe平衡(P<0.01),而在天府肉羊和成都麻羊中都保持了Hardy-Weinberg平衡(P>0.05)。
     对有产羔记录的山羊突变位点和胎次与它的产羔数进行最小二乘分析,结果表明:AA基因型天府肉羊头胎产羔数最小二乘均值分别比BB基因型多0.39只(P<0.05),第二胎、第三胎比头胎产羔数分别多0.6(P<0.05),0.87 P<0.01)。AA型基因型在成都麻羊中第三胎的产羔数最高为2.60只,AA型基因在三个品种山羊第二胎和第三胎(包括以上)的高产作用强于在头胎中的作用(P<0.05);AA基因型在各品种各胎次中的产羔数高于BB基因型(P<0.05。
     本研究中发现的1对引物与山羊的繁殖性状有关,但还需进一步研究。
Follicle-stimulating homone(FSH) is a pituitary glycoprotein hormone.FSH is ctitical hormones to follicular growth,development.,dominace,maturation and ovulation.They also influence the growth of the reproductive system and the quality of sperm.The polymorphisms of goat FSHβwas detected by PCR-SSCP method with different pairs of primers among 33 goats among 166 Tianfu goats,101 Boer Goats and 67 Chengdu Brown goats(Acquisited by Sichuan Agriculture University and Huiyuan Animal husbandry Company).In this study,Comparing the FSHβof sheep,bovine and pig in Genl3ank,the primers were designed and the 5' regulating region、the first and second exon of different goat breeds,are amplified.
     There are one SNP sites in the 5′regulating region of FSHβgene,Other two primers have no SNP site.Ii is A712G.T.The primers 1 have a middle PIC(0.25<PIC<0.5) in Tianfu goat.but have a lower PIC(PIC<0.25) in Boer Goat and Chengdu Brown goat.The result of x2 fitness test of primer 1 indicated that Boer Goat was hightly significantly biased from Hardy-Weinberg equilibrium(P<0.01),but Tianfu goat and Chengdu Brown goat are all kept in Hardy-Weinberg equilibrium(P>0.05).
     Among goats with littering records,the least squares analysis showed that:the litter of AA genotype in goat is more.0.39(P<0.05) than BB genotype,The litter of secong kidding in goat is more0.6 than that of first kidding in goat(P<0.05);The litter of third kidding in goat is more0.87 than that of first kidding in goat.(P<0.05).The litter is most in third kidding with AA genotype in Chengdu Brown goat.It is up to 2.60 per kidding.The effect of AA genotype in second and third kidding is stronger than that in first kidding.(P<0.05).The litter of AA genotype is more than BB genotype by same kidding of any kind of goats.
     Data obtainedni this expriment indicated that the polymorphism of fragments amplified by one pair of primer probably associate with goats productive trait,but need further studies.
