番茄红素的提取技术及稳定性研究

摘要
本文以新疆地产番茄酱为原料,研究了搅拌浸提、酶处理、微波提取等不同方法对番茄红素提取效果的影响,经研究确定了番茄红素的最适提取溶剂和最适提取工艺条件。通过对番茄红素油树脂脱溶方法的研究,采用气相色谱仪分析其脱溶效果,确定了高真空度是其安全脱溶的有效方法。研究了番茄红素纯化方法,确定了最适宜的结晶溶剂,认为结晶与重结晶相结合的方法是获得高纯度的番茄红素结晶的有效方法。以果蝇为对象,研究了番茄红素的抗衰老作用,证明番茄红素能显著延长果蝇的寿命。试验中采用紫外吸收光谱、薄层色谱及高效液相色谱等分析检测手段,对番茄红素粗提物、纯提物进行了定性、定量分析,并对各种处理条件下的稳定性进行了比较全面的研究,找出了影响番茄红素稳定性的各种因素,为番茄红素制品的护色和安全贮藏提供了参考和依据。
The paper used tomato sauce which was producted in XinJiang as raw material to study extraction efficiency of lycopene by microwave irradiation, mix extraction and enzyme treatment. By the experiment, we determined the optimum extracting solvent and technongical conditions. By studying the method of removing solvent in tomato oleoresin, efficiency of removing solvent was determined by gas chromatography(GC), we determined high vacuum conditions were the safety and efficiency method of removing solvent. We studied the way of purification of lycopene,determined the suitable solvent of crystallize, and regarded crystallization and recrystallization as ideal method which got high purification lycopene. We used Drosophila Melanag as objective of research to study antiaging effect of lycopene, the results showed that lycopene can obviously prolong Drosophila Melanag life span. In this study, ultraviolet spectrum, thin layer chromatography(TLC) and high performance liquid chromatography(HPLC) were used to identify the qualitative analysis and quantitative analysis of crude pigment of lycopene and pure pigment of lycopene, and studied the stability of lycopene in different conditions. finding out all kinds of facts which affect the stability of lycopene, and providing reference and evidence for protecting tincture and safety store of lycopene product.
