摘要
目的:建立蛇床子素壳寡糖纳米胶束中蛇床子素的含量测定方法。方法:采用高效液相色谱法测定蛇床子素壳寡糖纳米胶束中蛇床子素的含量:使用AgilentC18柱(4.6×200mm,5μm),流动相为甲醇-0.5%甲酸(75:25,v/v),检测波长322nm,进样量10μl,流速1.0ml/min,柱温25℃。结果:蛇床子素在0.78~50.00μg/ml的浓度范围内峰面积对浓度有良好的线性关系,且方法精密度良好;低、中、高3个浓度蛇床子素的回收率分别为101.7%、98.2%、98.9%,且RSD<2%。3批蛇床子素壳寡糖纳米胶束中蛇床子素的含量分别为154.0μg/ml、167.2μg/ml和175.1μg/ml。结论:本研究中高效液相色谱法测定蛇床子素聚合物胶束中蛇床子素含量的方法具有简便、快速、准确的优点,可用于蛇床子素壳寡糖纳米胶束中蛇床子素的含量测定。
Objective: To establish the method to determine the osthol content in osthol-chitosan nanomicelles. Methods: The content of osthol in osthol-chitosan nanomicelles was determined by high performance liquid chromatography by using Agilent C18 column(24.6×200 mm, 5μm), methanol-0.5% formic acid(75:25, v/v) as mobile phase, detection wavelength 322 nm, flow rate of 1.0 ml/min, and the column temperature was 25℃. Results: The linear range for osthol was in the range of 0.78~50.00μg/ml and the precision of the method was good. The recoveries of low, medium and high concentration osthol were 101.7%, 98.2% and 98.9% respectively, and the RSD was lower 2%. The osthol content in three batches osthol-chitosan nanomicelles were 154.0μg/ml, 167.2μg/ml and 175.1μg/ml. Conclusions: The method established in this study has the advantages of simple, rapid and accurate, and it can be used for determining the osthol content in ostholchitosan nanomicelles.
