基于中药整合药理学平台探究二陈汤防治肥胖的物质基础与作用机制

摘要
目的挖掘二陈汤防治肥胖的物质基础与作用机制。方法以中医药大数据的信息技术为手段,基于中药整合药理学计算平台的中药方剂数据库、中药材数据库、中药成分数据库、疾病症状靶标数据库为支撑,对二陈汤防治肥胖进行关联分析。共同构建"二陈汤-中药材-化学成分-核心靶标-关键通路"的可视化网络探讨二陈汤防治肥胖的物质基础和作用机制。结果利用平台筛选出二陈汤的305个化合物中有105个靶点与肥胖病的关键靶点相互关联,其中关键靶点HADHA、HADH、NSDHL等参与调控了磷脂酰胆碱生物合成、RNA聚合酶II启动子转录对内质网应激的正调控等过程。结论本实验通过中药整合药理学平台筛选出二陈汤化学成分,并预测出其治疗肥胖的作用靶点和相关通路,可能与甘油磷脂代谢、脂质代谢、神经系统方面密切相关。为阐明其防治肥胖的分子机制奠定基础,对深入挖掘和开发经典名方二陈汤有重要意义。
        OBJECTIVE To explore the potential mechanism of Erchen decoction for the prevention and treatment of obesity.METHODS With the big data of traditional Chinese medicine as a means of information technology,based on the traditional Chinese medicine prescription database,Chinese herbal medicine database,Chinese medicine component database and disease symptom target database of internet-based computation platform for integrative pharmacology of traditional Chinese medicine,we explore the possible mechanism of Erchen decoction in prevention and treatment of obesity. Establishing a visual multi-level network of Erchen decoctionChinese Herbs-Ingredients-Critical Targets and Pathways to explore the mechanism of Erchen decoction. RESULTS The one hundred and five targets of 305 compounds of Ershen decoction were screened out by platform to be correlated with key targets of obesity. The targets such as HADHA,HADH and NSDHL are involved in the regulation of phosphatidylcholine biosynthetic process,positive regulation of transcription from RNA polymerase Ⅱ promoter in response to endoplasmic reticulum stress,etc. CONCLUSION Through the platform that the mechanism of Erchen decoction in preventing and treating of obesity may be related to its role in glycerophospholipid metabolism,lipid metabolism,nervous system,etc were founded. in order to clarify its foundation for the molecular mechanism of obesity,and it is of great significance to dig deeper and develop the traditional Chinese medicine prescription Erchen decoction.
