摘要
为了有效提高矮紫杉种子萌发率,以矮紫杉种子为试验材料,采用L_9(3~4)正交试验,以GA_3、培养基、培养温度为正交试验的3因素,进行了种子的后熟处理,后取体胚接种于MS+AC 0.5 g·L~(-1)+琼脂7 g·L~(-1)+蔗糖30 g·L~(-1)上培养,研究后熟处理对矮紫杉种子体胚萌发的影响。结果表明:GA_3、培养基、培养温度对矮紫杉体胚的诱导影响显著,其中GA_3是主要因素,培养温度、培养基类型次之;矮紫杉体胚萌发前,种子后熟处理的最佳方式是将消毒好的种子接到MS+蔗糖30 g·L~(-1)+GA_3 1.0 mg·L~(-1)+加医用脱脂棉3.5 g·10 mL~(-1)的液体培养基中,置于白天23℃10 h、夜间13℃14 h的条件下培养,萌发率可达94.44%。
In order to explore the after-ripening on Taxus cuspidate cv.Nana seed embryo germination,the influence of body with Taxus Cuspidate cv.Nana seed as material,using L_9(3~4) orthogonal test table,with GA_3,culture medium,culture temperature for the orthogonal experiment of three factors,the seed cooked after processing,after take body embryo inoculation on MS+AC 0.5 g·L~(-1)+agar 7 g·L~(-1)+cultivated in sugar 30 g·L~(-1).The results showed that GA_3,culture medium and culture temperature had a significant influence on the induction of the embryo of the Taxus cuspidate cv.Nana,with GA_3 being the main factor,and the culture temperature and culture medium were secondary.Taxus cuspidate cv.Nana body after-ripened seeds before germination embryos processing was the best way to set up good seed disinfection MS+sugar 30 g·L~(-1),1.0 mg·L~(-1)+GA_3 and medical absorbent cotton 3.5 g·10 mL~(-1) of liquid medium,23 ℃ in the daytime 10 h,14 h 13 ℃ at night under the conditions of cultivation,germination rate can reach 94.44%.
