摘要
目的研究miRNA-145和miRNA-146a在缺血性脑卒中患者血清中的表达变化,探讨其与缺血性脑卒中的发生及发展的相关性。方法根据实验要求,选择2017年1月-2017年6月在我科住院治疗的缺血性脑卒中患者100例作为病例组,同期来我院门诊体检的40岁以上非缺血性脑卒中病史的健康体检者50例作为对照组。血液学检测的收集包括血清葡萄糖、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、同型半胱氨酸(HCY)、高密度脂蛋白胆固醇(HDL-C)等项目在内的研究资料。实验方法是荧光定量多聚核苷酸链式反应(实时荧光定量PCR)技术,对miRNA-145和miRNA-146a在病例组和对照组血清中的表达水平进行检测。收集好全部数据后进行统计分析。结果 (1)病例组血清中miRNA-145的表达水平要明显高于对照组血清中miRNA-145的表达水平(P <0. 05);(2)病例组血清中miRNA-146a的表达水平要明显低于对照组血清中miRNA-146a的表达水平(P> 0. 05)。结论 (1)缺血性脑卒中患者血清中miRNA-145的表达水平升高;(2)缺血性脑卒中患者血清中miRNA-146a的表达水平降低。
        Objective To investigate the expression changes of miRNA-145 and miRNA-146 a in the serum of patients with ischemic stroke,and to explore the correlation between them and the occurrence and development of ischemic stroke. Methods According to the relevant requirements of the experiment,we choose from January 2017 to June 2017 in the treatment of 100 cases of patients with ischemic stroke in our hospital as the case group,from January 2017 to June 2017 in our hospital outpatient examination over the age of 40 non ischemic stroke history of 50 cases of healthy persons as control group.The blood test collection data including serum glucose,serum total cholesterol( TC),serum triglyceride( TG),low density lipoprotein cholesterol( LDL-C),serum homocysteine( HCY),and high density lipoprotein cholesterol( HDL-C). The experimental method we use was fluorescence quantitative polymerase chain reaction( real-time quantitative PCR) technology,and detect the expression level of miRNA-145 and miRNA-146 a in case group and control group serum. After collecting all the data,the statistical analysis was carried out. Results( 1) The expression level of miRNA-145 in the serum of the case group was significantly higher than that of the miRNA-145 in the serum of the control group( P < 0. 05).( 2) The expression level of miRNA-146 a in the serum of the case group was significantly lower than that of the miRNA-146 a in the serum of the control group( P < 0. 05). Conclusions( 1) The level of miRNA-145 expression in the serum of ischemic stroke patients was increased.( 2) The level of miRNA-146 a expression in the serum of ischemic stroke patients decreased.
