摘要
目的研究锦州市啮齿动物中汉坦病毒(HV)的感染情况及基因分型。方法采用间接免疫荧光法(IFA)检测鼠肺中HV抗原;阳性样品扩增汉坦病毒M片段并测序;构建系统发生树进行系统发生分析及分型。结果 2011—2013年在锦州市出血热疫区,对捕获的977只鼠进行HV抗原检测。共检出HV抗原阳性鼠肺样品43份,病毒携带率为4. 40%。依据病毒分类及命名原则将43份样品分为7个毒株,每个毒株随机选取1份HV抗原阳性样品,对部分M片段进行扩增并测定序列。分析表明7株病毒与汉城病毒(SEOV)有较高的相似性,为95. 6%~100%,而与汉滩型病毒(HTNV)相似性仅为70. 2%~71. 4%,表明7份鼠肺均携带SEOV。结论锦州地区HV优势毒株为SEO型,其主要传染源是褐家鼠,系统进化树显示分离到7株SEO起源相同,均为S3亚型,并有明显的地区聚集性,变异较小,基因型别较为稳定。
Objective To study the epidemiology of hantavirus in rodents collected from Jinzhou. Methods Hantavirus antigens in the rat lungs were detected by immunofluorescence assay. Partial M segment sequences were amplified and then sequenced. The phylogenetic trees were constructed by maximum likelihood method to detect the genetic characteristics of hantavirus. Results From 2011 to 2013,a total of 977 host animals were captured.Hantavirus antigens detection showed 43 positive out of these rat lungs,accounting for 4. 40%. According to the classification of viruses and nomenclature,seven strains were separated from 43 positive samples,Partial M segment sequences were amplified with SEOV specific primers from 7 positive samples. The sequences held 95. 6%-100%similarity with SEOV,70. 2%-71. 4% with HTNV. Conclusion HV superiority strain was SEOV in Jinzhou area and the main host was Rattus norvegicus. Data from phylogenetic indicated that 7 separated viruses belonged to Seoul virus and the sub-genotype was SEOV-3.
