片仔癀对四氯化碳诱导肝纤维化大鼠肝功能及核因子-κB蛋白表达的影响

摘要
目的观察片仔癀对CCl_4诱导的肝纤维化大鼠肝功能和核因子-κB(NF-κB)蛋白表达的影响,探讨其作用机制。方法将24只大鼠随机分为正常对照组、模型组、片仔癀组、秋水仙碱组,每组6只,50%CCl_4灌胃8周制备肝纤维化模型。末次给药1 h后,血清测定ALT、AST、Ⅲ型前胶原、Ⅳ型胶原、层黏连蛋白和透明质酸含量,取肝脏计算肝指数,HE染色和VG染色观察肝组织病理形态,Western blot检测NF-κB蛋白表达。结果与正常对照组比较,模型组大鼠肝指数及ALT、AST、Ⅲ型前胶原、Ⅳ型胶原、层黏连蛋白、透明质酸含量和NF-κB蛋白表达明显升高(P<0.01);与模型组比较,各给药组大鼠肝指数及ALT、AST、Ⅲ型前胶原、Ⅳ型胶原、层黏连蛋白、透明质酸含量和NF-κB蛋白表达均明显降低(P<0.01)。结论片仔癀对CCl_4所致肝纤维化大鼠具有保护作用,其机制可能与抑制NF-κB的表达有关。
        Objective To investigate the protective effects of Pien Tze Huang on liver function and NF-κB protein expression in rats with hepatic fibrosis induced by CCl_4; To discuss its mechanism of action. Methods Totally 24 rats were randomly divided into four groups: normal group, model group, Pien Tze Huang group and colchicine group, with 6 rats in each group. The liver fibrosis model was prepared by intragastric administration of 50% CCl_4 for 8 weeks. After 1 hour of the last administration, serum was used to determine the contents of ALT, AST, type Ⅲprocollagen, type collagen, laminin and hyaluronic acid. Ⅳ Liver was taken to calculate the liver index. HE and VG staining were performed to observe the pathological damage. The expression level of NF-κB protein was determined by Western blot. Results Compared with the normal group, the levels of liver index, ALT, AST, type procollagen, Ⅲtype Ⅳ collagen, laminin, hyaluronic acid and NF-κB protein in the model group significantly increased(P<0.01). Compared with the model group, the liver index, ALT, AST, type Ⅲprocollagen, type Ⅳcollagen, laminin, hyaluronic acid and NF-κB protein significantly decreased in each administration group(P<0.01, P<0.05). Conclusion Pien Tze Huang has a significant protective effect on hepatic fibrosis induced by CCl_4, which may be related to the inhibition of NF-κB expression.
